| Objective:To find genes related to fibroblast proliferation in Cherubism by comparing gene expression profile in RAW 264.7 cells with overexpressed wildtype and mutant SH3BP2 gene.Methods:Total RNA in RAW 264.7 cells with overexpressed wildtype or mutant SH3BP2 gene were extracted respectively by Trizol and analyzed by microarray to compare differences in gene expression profiles, followed by database searching to target most relevant genes and realtime PCR to verify the most significant related genes in fibroblast proliferation in Cherubism.Results:Microarray data indicated that 734 genes show significant expression differences between the two cell lines, among which,313 genes are up-regulated and 421 down-regulated in cells with mutant SH3BP2 compared with wildtype. Following investigation with database searching and realtime PCR confirmed 4 genes that are top relevant to fibroblast proliferation:CXCL10, IL10, Tnfrsfl3b and Pf4.Conclusion:Results from our study showed expression profiles differences between the two cell lines. We pointed out 4 main genes which could be the potential key components in the mechanism of fibroblast proliferation in the context of Cherubism, and provide us with a bright new window into further study of the disease. |
