Objective:During the process of aging, the incidence of myocardial ischemia (MI) is gradually rising.MI is the initial stage of a variety of cardiovascular disease or disorder, which’s major clinical symptoms are angina pectoris or myocardial infarction. MI belongs to the category of "Chest pain", "heartache" in Traditional Chinese Medicine(TCM). Due to MI is a severe disease with rapid occurance and high mortality but poor in prevention and treatment, it’s great significance to study on the TCM research of preventing and treating MI. Based on our team’s previous studies on Zuogui Pill (ZGP) regulates the proliferation and differentiation of aging rats Bone-marrow Mesenchymal Stem Cells (MSC) and according to the hypothesis of "Kidney essence deficiency leads to aging theory" and "stem cells have the same properties as congenital essence and is a existence form in the cellular level", we further explore ZGP regulate the MSC of aging-MI^ model rats migrating to the injury site of heart tissue to repaire. The study will further defining scientificity of the hypothesis and provide experimental evidence for treatment of cardiovascular diseases by Chinese medicineMethods:1. By intraperitoneal injection of D-galactose to establish subacute aging rats model. Gavage treatment with ZGP solution.2. By intraperitoneal injection of isoproterenol to establish rat model of myocardial ischemia3. Blood tests in each group; serum level tests of SOD activity, MDA concentration to evaluate aging model rats;Test the electrocardiogram ST segment to evaluate rat model of myocardial ischemia.4. Detection of bone marrow and blood concentrations of SCF;detection of bone marrow and blood CD105,CD117 positive cells by flow cytometry,5. Test the Serum level of LDH activity, CK activity, Hematoxylin-eosin staining of cardiac tissue to evaluate the rats’cardiac injury level and the TNF-a concentration, IL-6 concentration; test the cardiac tissue of SOD activity, MDA concentration, TNF-α concentrations, IL-6 concentration; Observation of cardiac tissue immunofluorescence of CD105, CD117-positive cells and test the VWF, connexin-43 expression to evaluate MSC homing ability to repair the cardiac injuryResults:1. Blood tests:compared with the aging+MI group, the number of white blood cells of the aging+MI+ZGP group significantly decreased(p<0.05)2. SOD and MDA tests:the SOD activity of the aging group was significantly lower than the normal group while the MDA concentration was significantly higher than the normal group (p<0.05); compared with the aging+MI group, the SOD activity of the aging+MI+ZGP group significantly increased while the MDA concentration significantly decreased(p<0.05)3. The electrocardiogram ST segment:compared with the aging+MI group, the aging+MI+ZGP group significantly changed typically.4. Bone marrow and blood concentrations of SCF:compared with the aging+MI group, both bone marrow and blood concentrations of SCF of the aging+MI+ZGP group significantly increased (p<0.05)5. Serum level of LDH activity, CK activity:compared with the aging+MI group, the aging+MI+ZGP group significantly increased(p<0.05); compared with the aging group, the aging+MI group significantly increased(p<0.05); Serum level of TNF-α concentration, IL-6 concentration:compared with the aging+MI group, the aging+MI+ZGP group significantly decreased(p<0.05; p<0.01);The cardiac tissue of SOD activity, MDA concentration, TNF-α concentrations, IL-6 concentration:compared with the aging+MI group, The SOD activity of the aging+MI+ZGP group significantly increased while the MDA concentration significantly decreased (p<0.05); compared with the aging+MI group, the TNF-α concentrations and IL-6 concentration of the aging+MI+ZGP group significantly decreased(p<0.05 p<0.01); Hematoxylin-eosin staining of cardiac tissue showed that the aging+MI group with fiber disorder, cytoplasm edema while the aging+MI+ZGP group improved significantly.Immunofluorescence of CD105, CD117-positive cells:compared with the aging+MI group. The SOD activity of the aging+MI+ZGP group significantly increased (p<0.05 p<0.01); The VWF connexin-43 expression:compared with the aging+MI group, the VWF expression of the aging+MI+ZGP group significantly decreased while the connexin-43 expression significantly increased (p<0.05).Conclusions:1.succeed by intraperitoneal injection of D- galactose and isoproterenol in establishing aging rat model of MI2. ZGP can regulatethe bone marrowCD105 and CD117-positive cells torepaircardiactissue damage;3. The machanism that ZGP regulate MSC maybe related to the SCF increasing.4. This-study proved the "stem cells have the same properties as congenital essence and is a existence form in the cellular level" hypothesis of science in the aspect of disease to rich "kidney essence" new substance of modern theory.
