| Objective:In order to investigate the association between the ABCC2 C-24T, G1249A and C3972T polymorphism with both response and adverse drug reaction to antiepileptic drugs.Methods:We searched relevant articles in PUBMED and Web of Science. Dominant, recessive, co-dominant and allele genetic models were tried to perform for the three genetic polymorphisms. The ORs and their 95%CI, the publication bias and the results of influence analysis were observed in all the meta-analysis.Results:Totally,14 previous publications with 4435 patients were finally included. In the overall analysis of drug response, we found that TT/CT+CC and TT/CC genetic models of ABCC2 C-24T were associated with response to antiepileptic drugs (P< 0.05 and OR>1), but ABCC2 G1249A and C3972T did not show similar results. Moreover, we could not obtain any significance between ABCC2 polymorphisms and the drug response in subgroup analysis (Asian and non-Asian, respectively). Moreover, data showed that ABCC2 G1249A was associated with response to antiepileptic drugs in Chinese of GA+AA/GG, GA/GG and A/G models with P-value of 0.001,0.000 and 0.001, respectively. But we could not find similar results about ABCC2 G1249A and C3972T in Chinese (all P> 0.05). Meanwhile, we failed to demonstrate monotherapy of Carbamazepine/Oxcarbazepine in an important association with ABCC2 polymorphisms according to the eligible data. In the part of adverse drug reaction for antiepileptic drugs, ABCC2 G1249A was observed in a significant relation with antiepileptic drugs in GG/GA+AA, GG/GA and G/A genotypes with all P< 0.01.Conclusions:ABCC2 C-24T was associated with drug response to antiepileptic drugs and ABCC2 G1249A was significantly associated with adverse drug reaction to antiepileptic drugs in the overall analysis. Besides, it was notable that ABCC2 G1249A was in a significant association with drug response to antiepileptic drugs in Chinese. |
