Objective:To study the biocompatibility of the absorbable polylactic acid (PLA) root post film prepared by solvent evaporation film.Methods:The PLA root post films and its leaching liquor were prepared by the method of solvent evaporation. The biocompatibility of PLA films were measured through both vitro and vivo tests. A. vitro test:cultivated in vitro, the sources of HGFs were identified initially by the immunohistochemistry method. Incubated in PLA post film leaching liquor, the toxicity reaction, the survival rate and the morphological change of the HGFs were observed and tested by morphological observation, MTT assay and flow cytometry. The HGFs were implanted on the PLA post film and observed by DAPI staining. B. vivo test: After being implanted the PLA post film, the surrounding subcutaneous tissue of SD rats and the implants were removed from the rats’ back in the 1th,4th,8th, 12th week to observe the incisions’ general condition. And the organization was given the HE stain to find the organization’s condition of local tissue inflammation and foreign body reaction through the infiltration degree of inflammatory fibroblasts.Results:The effects of the PLA post film leaching liquor on proliferation of HGFs and fibroblasts toxicity was not significant. The living fibroblasts rate was similar to the normal control group. The HGFs was able to grow on the PLA film.The results of rats’organization general observation and HE stained were similar to the control group which met all the demand made by biological safety.Conclusion:PLA absorbable root post film prepared by solvent evaporation film has good biocompatibility.
