| Objective To explore the effect of Resveratrol on the proliferation and metastasis of human gastric cancer cell line SGC-7901 and the expression of p-ERK, CXCR4, CXCR7,VEGF,MMP-9.Methods The gastric cell line SGC-7901 was treated with CXCL12 and Resv, the proliferation of SGC-7901 cells was tested by MTT. The migration and invasion rate was tested by Wound healing assay and Tranwell Chambers respectively. The levels of CXCR4, CXCR7, VEGF and MMP-9 mRNA were measured by Reverse Transcription Polymerase Chain Reaction (RT-PCR). The phosphorylation level of ERK protein was measured by Western Blot.Results Different concentrations of CXCL12 promoted the proliferation of gastric cells. CXCL12 could significantly promote the migration and invasion of gastric cells. CXCL12 also up regulated the protein of p-ERK and the mRNA of CXCR4, CXCR7, VEGF and MMP-9. On the other hand, Resveratrol could inhibit the CXCL12-induced proliferation, migration and invasion of gastric cells, and down-regulated the translation of p-ERK and transcription of CXCR4, CXCR7, VEGF and MMP-9.Conclusion Resveratrol could inhibit the proliferation, migration, invasion of gastric cancer cell. The mechanism may be related with decreasing the expression of p-ERK, CXCR4, CXCR7, VEGF and MMP-9. |
