ObjectiveTo illustrate the effect of CXCL12 on the proliferation,invasion and migration of cervical cancer Caski cells,analyze the underlying mechanism.MethodsThe human cervical cancer Caski cells line cultured in vitro were divided into control group;CXCL12 groups(25 ng / ml,50 ng / ml,100 ng / ml,200 ng / ml);CXCL12 + PD98059 group(100 ng / ml CXCL12 + 100 ?mol / L PD98059);PD98059 group(100 ?mol / L).The proliferation,migration and invasion ability of the cells were detected by MTT,cell scratch test,transwell invasion test and cell adhesion test.The expression of ERK,p-ERK,MMP-2,ETS-1and uPA proteins were detected by Western blot.ResultsThe research shows that CXCL12 concentration-dependently promoted cell proliferation,adhesion,scratch healing,cell invasion(P < 0.01);The difference between 100 ng/ml and 200 ng/ml was not staistically significant(P > 0.05).PD98059 has the capcity to block the above effects of CXCL12.Comparing with the control group,CXCL12 promoted the expression of p-ERK,ETS-1,MMP-2,uPA proteins(P < 0.01)significantly.Comparing with the CXCL12 group,PD98059 significantly decreased the above protein expressionConclusionThe proliferation,invasion and migration of cervical cancer cells can be accelerated due to the reason that CXCL12 could highlight the expression of ETS-1,MMP-2 and uPA proteins,which is contributed to the possibility of increasing ERK signaling pathway.
