Objective To investigate the effect of Shendibushen Capsule on 24-hour urinary protein excretion of UUO rats and the expression of p-JAK2 and p-STAT3 to explore the mechanism of anti renal interstitial fibrosis.Methods 80 Healthy male SPF SD rats, randomly divided them into:control group,sham operation group,UUO group,Niaoduqing group,Shendibushen group.Using UUO induced renal interstitial fibrosis model.the blank group,sham operation group,UUO group were given 2ml/d normal saline irrigation and Niaoduqing group according to the 2.25g/(kg·d) preparation of mixed suspension gavage,Shendibushen group according to the 0.324g/(kg·d) mixed suspension gavage.It last 4 weeks.For the preparation of Model 2 week and 4-week time point to collect specimens,observe the general condition and body weight of rats,and 24-hour urinary protein changes,renal histological changes,the renal tissue p-JAK2、p-STAT3 expression at each time point.Results Blank group and sham operation group compared with the observed indicators were not statistically significant( P >0.05).24-hour urinary protein excretion in each group was not statistical significant( P >0.05).all the observed indexes in UUO group were compared with those in the blank group and sham operation group( P <0.05).After treatment,Shendibushen group rats general state were obviously superior to the UUO group at the same time,the weight gain was statistically significant( P <0.05);pathological changes compared with the UUO group significantly reduced.Shendibushen capsule can inhibit renal tissue p-JAK2、p-STAT3 protein expression with the same period in UUO group was statistically significant( P <0.05).There was statistical significant( P <0.05) in the Shendibushen group.Shendibushen group with reference to the same period Niaoduqing group showed no statistical significance( P >0.05).Conclusion1、Shendibushen capsule can improve RIF rats general status.2、Shendibushen capsule can reduce the pathological changes in rat kidney RIF3、Shendibushen capsule may through down the expression of p-JAK2 and p-STAT3 protein in renal tissue to prevent renal interstitial fibrosis. |
