Objective:This paper aims to observe the effects of acupoint catgut-embedding(ACE) at "Zusanli" on fat reduction in diet-induced-obesity mice, and the expression of protein in the liver, as the experimental evidence of acupoint catgut-embedding to lose weight.Method:We choose 30 healthy and clean weaned C57BL/6J male mice to adaptive feeding one week, and then randomly divide them into normal group and high-fat group, the normal group with normal diet, high-fat group with high fat diet, so as to establish food borne obese mice. After successful modeling, the obese mice are randomly divided into ACE group and model group. Treatment methods:The ACE group is fixed on the self-made fixator,75% alcohol disinfect both sides of "Zusanli", No.7 with acupuncture at "Zusanli", penetrate into the depth of 4mm. According to the corresponding point of muscle will appropriate length into the tissue,after acupuncture out, press the acupuncture hole for a moment. According to the absorption rate of catgut treatment, once every 10 days for 3 times. The normal group and the model group:both groups are fixed on the self-made fixator without any treatment,as the same fixed time with the ACE group. The weight of mice in each group were recorded during the treatment period. After the end of the treatment, the serum, fat and liver were collected, and the differences of body weight, body fat, blood glucose, blood lipid and other related indexes in each group were compared. The level of total cholesterol and triglyceride in liver tissue was detected. Compare the fat morphological differences between groups of mice with HE staining. The morphological differences of the livers of the mice were observed and compared by oil red staining. The expression level of FAS、PERILIPIN、C/EBP a protein in liver was observed by Western-blot.Result:Catgut embedding therapy on body weight of obese mice significantly decreased:before the treatment the weight of obese mice (model group and ACE group) was significantly higher than that in normal group, the difference was statistically significant (P< 0.01). After treatment, the ACE group, body weight were significantly decreased, the weight and weight reducing rate compared with the other two groups, the difference was statistically significant (P< 0.01).Catgut embedding therapy on body fat in obese mice decreased significantly:high fat diet induced model mice inguinal, epididymal fat weight were higher than that of the normal mice, the difference is statistically significant (P< 0.01); after treatment, ACE group mice inguinal, epididymal fat weight than those in the model group significantly decreased, the difference is statistically significant (P< 0.01). After brown fat volume was significantly lower than that in the model group decreased, the difference is statistically significant (P< 0.05).Mice in the model group,TC、LDL、Glu levels than those of normal mice was elevated, the difference is statistically significant (P< 0.05); after treatment, the ACE group mice TC-. LDL levels compared with the model group significantly reduced, the difference is statistically significant (P< 0.01), TG and Glu levels compared with the model group was reduced, the difference was statistically significant (P<0.05).Compared with the normal group,TC in model group mice liver homogenate was increased significantly (P< 0.01), TG level increased (P< 0.05). In HE stain showed fat cell sizesignificantly increased, cytoplasmic lipid droplets larger; liver oil red staining display liver cytoplasmic lipid droplets dyed red by oil red, and distributed in the cytoplasm; Fas、perilipin、 C/EBP alpha protein in liver tissue expression on the high side.Compared with the model group,TG in ACE group liver homogenate decreased significantly (P< 0.01), TC levels decreased (P< 0.05); fat cell size is uniform, the volume of cells was significantly decreased, cytoplasmic lipid droplets are smaller; liver oil red staining of the lipid drop obviously reduced;the FAS% perilipin, C/EBP alpha protein in liver tissue expression is low.Conclusion:Acupoint catgut-embedding "Zusanli" can reduce the content of food borne obesity mice weight, visceral fat weight, blood glucose, blood lipid level, hepatic lipid. Inhibition of FAS、 Perilipin、C/EBP alpha protein expression in liver tissues of obese mice, thereby at the root of the inhibition of the disintegration and generation of fat, to promote the decomposition of fat. This method have good effect on weight loss to food borne obese mice.
