In theory research, this article has carried on the deep discussion on the etiology and pathogenesis of Alzheimer disease (AD) in the field of Chinese medicine from the clinical manifestation of AD patients, especially the old people and senile dementia patients’physiological and pathological characteristics. The dysfunction of liver and spleen was considered closely related to the origin and symptom of AD in the perspective of liver depression and spleen deficiency. It will blinded your sanity directly and eventually lead to dementia, when the liver Qi stagnation syndrome happens and the liver disorders. As a saying goes, all ills and diseases ascribed to the liver stagnation, liver is the core power for Qi activities. If the liver disorders, it will impede the running of Qi and blood. Because it causes Qifen firstly, then the Qi of whole body sparse release is abnormal, it hurts the blood system with time. The Qi was unable to drive the blood flow and finally made the blood stasis block, venation estrangement, it may cause the disease that we called stroke. Also, it will make your body fluid convert to sputum, and move up to affect the brain. There were some researches conclude that the sputum and silt caused by the liver depression, will develop into the depression, which can make medical conditions worse with time. The traditional Chinese medicine theory thinks, if the pathological result of this kind of phlegm and blood stasis block the brain, it can cause cerebral infarction, which was recognized as one of the factors of vascular dementia known by the public. The liver depression also may trigger the dysfunction of spleen, which can reject the conversion from food and water to energy.When the sea of marrow loss of curing, the dementia develops. In addition, the dysfunction of spleen can produce harmful pathogenic products, such as phlegm turbidity, which was one of the important causes of dementia. According to the progress of AD in western medicine, it can conclude that the soluble AP is closely related to the severity of AD in one’s brain, the liver is the main organ to clear Aβ away, which can protect one’s brain. So, invigorating liver is one of the important methods in the treatment of AD. Western medicine on AD liver’s "set a system to eliminate Aβ theory has the same effect as the Chinese medicine dementia’s "liver and spleen imbalance cause the damage of brain" etiology and pathogenesis and the "regulating liver and tonifying spleen" treatment principle.In the formal experimental research, AD models were constructed by the combined application of the injection of Pentobarbital Sodium in abdomen and 3 μL Aβ solution in cranial cavity. This building method can mould an animal dementia model for a long time, which has been widely used in the experiment of senile dementia modeling. Experimental subjects were divided into four categories, control group, model group,18-α Diammonium Glycyrrhizinate low and high dosage groups. The experiment mainly studied on the function of 18-α Diammonium Glycyrrhizinate forAD from the changes in behavior, pathological section, gene expression. This experiment contents the senile dementia rats water maze test, ELISA detection, Western blot protein detection and Immunofluorescence detection concluded by statistics processing. The experimental results show that:18-α Diammonium Glycyrrhizinate can effectively improve rats learning and memory ability and the high dose group improved more obviously, high doses of glycyrrhizic acid treatment can obviously increase the brain microvascular endothelial cells expressing LRP1 protein, lower the levels of the brain Aβ to play an important role in the treatment of AD.
