Senile dementia (Alzhemier’s disease, AD) is a sort of common neurodegenerative disease, which is characterized by cognitive impairment, mental and behavior disorder and social withdrawal. The effective therapeutic method is lack because of indefinite etiology pathogenesis. Traditional Chinese Medicine holds that this disease is root deficiency and branch excess, which is caused by deficiency of liver and kidney, insufficiency of marrow-sea, heart-spleen deficiency, qi stagnation, blood stasis and sputum.This article discusses the experimental therapeutic effect of Modified Sanjiasan decoction on Senile dementia in various theoretical and experimental levels. In the theoretical study aspect, we retrospect the interrelated thesis of Senile dementia in TCM ancient books and modern medicine and prophase studies of Modified Sanjiasan decoction on Senile dementia.In the experimental study aspect, we take BV2 cells and PC 12 cells as research objects, BV2 cells are divided into follow groups:normal control group(A), model group(B), Modified Sanjiasan decoction high dose group(C), Modified Sanjiasan decoction middle dose group(D), Modified Sanjiasan decoction low dose group(E) and Huperzine group(F).When cells adhering to the wall, we add Modified Sanjiasan decoction-contained cerebrospinal fluid and Huperzine-contained cerebrospinal fluid into C< D、E and F groups, incubate for 2 hours, then we add LPS with final concentration of 1 ug/ml into B、C、D、E and F groups, the culture supernatant were collected and cell proteins were extracted after incubated for 24 hours. Detect the different groups BV2 cells secreted expression of NO by Griess method, the secreted expression of iNOS、IL-1β and TNF-α by ELISA and the expression of JNK and p-JNK by Western blot. Then we take the culture supernatant as conditioned medium to cultivate PC 12 cells, the cell proteins were extracted after incubated for 24 hours, then detect the expression of APP and BACE by Western blot.The results indicate that the secreted expression of NO、iNOS、IL-1β and TNF-a and the expression of p-JNK of model group were increased, the secreted expression of NO、iNOS、IL-1β、and TNF-α and the expression of p-JNK of Modified Sanjiasan decoction high dose group and Modified Sanjiasan decoction middle dose group were declined. The expression of APP of Modified Sanjiasan decoction high dose group, Modified Sanjiasan decoction middle dose group and Modified Sanjiasan decoction low dose group were declined compared with model group. The expression of BACE of Modified Sanjiasan decoction middle dose group and Modified Sanjiasan decoction low dose group were declined compared with model group. The results suggest that Modified Sanjiasan decoction may have a positive therapeutic effect on Senile dementia, the mechanism of this action may be concerned with regulating the expression of p-JNK and decreasing the generation of inflammatory medium, thus alleviate the BV2 microglia’s toxic effect on PC 12 cells.
