Objective:To investigate the role of ginsenoside Rbl on isolated-heart with myocardial ischemia-reperfusion (I/R) injury in rat.Methods:48 SD rats were divided into 6 groups(6 rats in each group):control group (K-H buffer perfused 130 min), I/R group (K-H buffer perfused 40 min, Ischemia 30min,K-H buffer reperfused 60min), ginsenoside Rbl (1,5,10.20μM) groups (K-H buffer perfused 10 min, G-Rbl K-H buffer perfused 30min, ischemia 30min, K-H buffer reperfused 60min). The Langendorff experimental installation was used to establish the isolated-heart perfusion and myocardial I/R.Left ventricular developed pressure (LVDP), changes in the rate of left ventricular pressure (LV dP/dP), heart rate (HR)and other cardiac function indexs were monitored during the process; the indicators of creatine kinases, troponin and myoglobin were tested to assessment of myocardial injury degrees; the myocardial infarct size was detected with TTC staining; the myocardial structure injury was detected with HE staining.Results:The Langendorff experimental installation was used to establish the isolated-heart perfusion and myocardial I/R, the development of left ventricular pressure (LVDP), left ventricular pressure change rate (LV dp/dt) and heart rate (HR) for real-time monitoring and recording, the results showed:Comparing with control group, I/R group’s LVDP significantly reduced (P<0.01), both myocardial infarction and enzymes and other indicators and structure injury have significantly increased (P<0.01)in I/R group.Comparing with I/R group, Rb1(1,5,10,20 μM)pretreatment increased LVDP and reduced myocardial infarction size and mycardical enzymes (P<0.05)Conclusions:The Langendorff experimental installation was used to establish the isolated-heart perfusion in a certain period of time does not affect cardiac electrophysiology index, won’t cause myocardial ischemia, myocardial injury and myocardial structure change phenomenon;Ginsenoside Rbl can effectively improve the LVDP, decrease infarct size,and indicators of creatine kinase and troponin after myocardial I/R injury to protecting myocardial ischemia-reperfusion injury.
