Objective: The establishment of integral experiment of human hepatoma SMMC-7221 cell line in vitro nude mouse animal model is aimed at investigating the tumor suppressor and radiosensitivity effects of Docetaxel on human hepatoma cell line SMMC-7221. The anti-angiogenic effect of Docetaxel is observed through the positive expression level of VEGF protein. The relativity between expression level of VEGF protein and the radiosensitivity effect of Docetaxel is analyzed through the testing of the expression level of VEGF protein, as well as the observation of the anti-angiogenic effect of Docetaxel. And continually study the relationship between the radiosensitivity effect of Docetaxel and the tumor blood vessels regulation.Methods:Human hepatoma SMMC-7221 cell line as the object of the research to establish nude mouse animal model:1. The volumes and weights of nude mice subcutaneous tumor were monitored, and the growth curve was drew.2. The morphological and histological characteristics of nude mice subcutaneous tumor was observed by HE staining.3. The apoptpsis of subcutaneous tumor in nude mice was detected by TUNEL.4. The use of immunohistochemical method was applied to test the expression of VEGF in nude mice tumor.Results:1. The nude mice tumor volume and tumor weight in the simple Docetaxel group, the Gamma Knife irradiation group, and the Docetaxel combined with Gamma knife irradiation group was lower than that in the control group. Furthermore; the tumor growth inhibition was most obvious in the Docetaxel combined with Gamma knife irradiation group, and the sensitivity coefficient of Docetaxel was 1.55.2. In these four treatment groups, the degree of the shrinkage, density decreasing, the cancer nest formation and tumor cells necrosis in tumor cells could show the following rankings:Docetaxel and Gamma knife integrated irradiation group ranks the first, seconded the simple Docetaxel group, followed by the simple Gamma knife irradiation group, and final the control group.3. In the simple Gamma knife irradiation group, the expression level of VEGF protein in the liver cancer cell was increased compared to the control group. To the contrary, the expression level of VEGF protein was decreased in the simple Docetaxel group. And the expression level of VEGF protein in the Docetaxel and Gamma knife irradiation group had the lowest among them.Conclusion:1. It could be revealed that Docetaxel showed a significant tumor suppressor effect on the hepatocellular carcinoma SMMC-7221 cell line. The integrated application of Docetaxel and Gamma knife irradiation could enhance the tumor suppressor effect. Docetaxel could improve the sensitivity of hepatocellular carcinoma cells to radiation.2. Docetaxel could level down the expression level of VEGF protein to inhibit the formation of blood vessels, proving that the tumor suppressor effect of Docetaxel on hepatoma cell would be correlated with the reducing of newborn blood vessels formation in the tumor.3. Docetaxel combined with Gamma knife irradiation could show a demonstrable reduction in the expression level of VEGF protein, and the observable inhibition of angiogenesis, which would suggest that the correlation between Docetaxel radiosensitization effect and inhibition of angiogenesis in hepatocellular carcinoma exists.
