| Objective To detect the expression of EP1 and COX-2 in HCC and adjacent tissues and explore the correlation between EP1 and COX-2 and the prognosis of HCC, to provide a new index for judging the prognosis of patients with HCC.Methods 1. In 2013,we collect 48 cases of fresh specimens with radical hepatic resection of HCC patients in the Affiliated Tumor Hospital of Guangxi Medical University.By polymerase chain reaction (polymerase chain reaction (PCR) and WB (WB, WB) method,we detect the expression of COX-2 and EP1 in high differentiation and low differentiation HCC group.2. In 2008-2011,we collect 116 cases of paraffin embedded specimens with radical hepatic resection of HCC patients in the Affiliated Tumor Hospital of Guangxi Medical University. According to IHC score, COX-2 or EP1 were divided into two groups.We compared the difference of free survival and (DFS) and Overall (survival OS, Disease) in patients with low or high COX-2 or EP1 group.Result 1. COX-2 mRNA is highly expressed in high differentiation group, while it is lowly expressed in low differentiation group (P=0.051). EPimRNA is highly expressed in low differentiation group, while it is lowly expressed in high differentiation group(P=0.087). COX-2 protein is highly expressed in high differentiation group, while it is lowly expressed in low differentiation group (P=0.003). EP1 protein is highly expressed in low differentiation group, while it is lowly expressed in high differentiation group(P=0.001)2. Univariate analysis showed that the AFP>400ng/mL,incomplete capsule,tumor diameter greater than or equal to 5 cm,Edmondson gradeâ…¢-â…£ and EP1 expression affected overall survival. Multivariate analysis indicated that AFP>400ng/mL, tumor diameter greater than or equal to 5 cm and the high expression of EP1 is an independent risk factors of the overall survival. Univariate analysis showed that the AFP>400ng/mL, incomplete capsule, tumor diameter is equal to or more than 5 cm, the high expression of COX-2 affected DFS. Multivariate analysis indicated that incomplete capsule, tumor diameter is equal to or more than 5 cm, the high expression of COX-2 were independent risk factors for DFS.Conclution 1. The expression of COX-2 is high in highly differentiated HCC tissues and the expression of COX-2 is low in low differentiated HCC tissues. The expression of EP1 is high in low differentiated HCC tissues and the expression of EP1 is high in low differentiated HCC tissues.2. OS is better in low expression group of EP1 than in high expression group of EP1. DFS is better in low expression group of COX-2 than in high expression group of COX-2. |
