Objective: Investigating the expression of PED / PEA-15 gene in pancreatic cancer, Exploring the relationship with PED / PEA-15 gene expression levels and after radical operation of overall survival(OS) of pancreatic cancer patients and its clinical values, designing and producing the specific siRNA of PED / PEA-15 gene and then transfecting into the human pancreatic cancer cell line PANC-1 to silence PED / PEA-15 gene expression in PANC-1 cells, To investigate the relationship with the expression of PED / PEA-15 gene and its influence of pancreatic cancer cell proliferation, migration and invasion ability.Method: 1. Using real time PCR, immunohistochemistry and Western blot to detect the expression of PED/ PEA-15 in pancreatic cancer and corresponding adjacent noncancerous tissues. 2. Collect clinical and pathological data of PED / PEA-15 in pancreatic cancer, and then analyze the relationship between the expression level of PED/ PEA-15 and patients survival time. 3.Down regulating the expression of PED / PEA-15 in pancreatic cancer cell PANC-1, and then detecting the effection on the PANC-1 cell in proliferation, invasion and migration ability.Results: By the methods of immunohistochemistry, RT-PCR, Western blot,these showed that the expression of PED/PEA-15 in pancreatic cancer were higher than those of corresponding adjacent tissues(P < 0.05), it suggests that it may play the cancer promoting effects in the progression of pancreatic cancer; By detecting the expression level of PED/PEA-15 in pancreatic carcinoma patients’ tissues, and following up the 5 years survival time of patients, the Kaplan-Meier curves showed that the expression of PED/PEA-15 with high level of the total survival time after radical resection was significantly lower than patients with low levels of expression(P < 0.05), and the statistic analysis showed that the age and gender, tumor size, invasion depth had no significant relationship with the expression level of PED/PEA-15. But the lymph node metastasis and the degree of tumor differentiation, TNM staging, metastasis has significant relationship with the expression level of PED/PEA-15(P < 0.05); Down regulation the expression of PED / PEA-15 in PANC-1 cell line and then detecting the proliferation, invasion, migration ability.It shows that the PED/ PEA-15 expression levels has positive correlation with pancreatic cancer cell proliferation, migration and invasion ability(P < 0.05).Conclusion: The expression of PED/ PEA-15 in pancreatic cancer tissues was significantly higher than that of corresponding paracancerous tissues and normal pancreatic tissue; In human pancreatic cancer cell line PANC-1, AsPC-1 and BXPC-3, CFPAC-1, SW1990, the expression of PED/ PEA-15 was significantly higher than that of the normal epithelial cell HEK293; The expression lever of PED/ PEA-15 has high value to assess the influence of the prognosis of patients with pancreatic cancer surgery after 5-year overall survival time, it can also provides reference opinions for risk classification of patients with pancreatic cancer and formulating a reasonable therapy; using the specific siRNA for PED/ PEA-15 gene and transfecting into PANC-1 cells can inhibit the proliferation activity of pancreatic cancer PANC-1 cell and induce the migration and invasion ability, PED / PEA-15 plays an important weight in the development of tumors.
