Objective: To detect the protein expression level of Forkhead box C1(FOXC1) in 5-8F cell line and investigate the influence on the biological behavior of 5-8F cell line of nasopharyngeal carcinoma(NPC)after FOXC1 knocked down. Methods: The protein expression level of FOXC1 in the human immortalized nasopharyngeal cell line NP69 and the nasopharyngeal carcinoma cell line with high metastatic potential5-8F was detected by Western blotting; The small interfering RNA(siRNA) was used to interfer the expression of FOXC1 in the 5-8F cell line, and the efficiency of interference was detected by Western blotting. The change of the migration ability of 5-8F cell line was observed by scratch test, both the apoptosis and the cell cycle of 5-8F were analyzed by the flow cytometry after FOXC1 knocked down in 5-8Fcell line. The protein expression levels of Fibonectin, Vimentin, Bcl-2and Cyclin D1 in 5-8F cell line after FOXC1 knocked down were detected by Western blotting. Results: The protein expression level of FOXC1 in5-8F cell line was much higher than the protein expression level of FOXC1 in NP69 cell line, and the difference between them was statistically significant(P<0.05). Compared with the groups of controls,the ability of cell migration significantly got down after FOXC1 knocked down in the 5-8F cell line(P <0.05); the apoptosis in 5-8F cell line was increased(P <0.05), and the cell cycle of 5-8F cell line was increased in Synthesis stage(P <0.05), and the protein expression levels of Fibonectin,Vimentin, Bcl-2 and CyclinD1 in 5-8F cell line after FOXC1 knocked down were decreased. Conclusion: The migration ability of in 5-8F cell line can be inhibited, the apoptosis can be promoted and the cell cycle distribution can be influenced by down-regulation the expression of FOXC1 in 5-8F cell line.
