Objective To investigate the inhibitory effect of hTRAIL on synovial proliferation in rats with collagen induced arthritis. Methods36 wistar rats were randomly divided into six groups, the control(group1), control 1μg hTRAIL–treated(group2), model group(group3), 1μg hTRAIL–treated(group4), 1.5μg hTRAIL–treated group(group5), 2μg hTRAIL–treated(group6), To create CIA model, complete Freund’s adjuvant and TypeⅡ collagen were injected into rats. After creating CIA model successfully, the 4 groups were given different doses of hTRAIL. Ankle joint diameter and swelling rate were adopted to evaluate the inflammatory reaction in rats in the CIA 16 d, 20 d and 24 d. HE staining was used to observe the joint pathological morphology under an inverted electron microscope. The synovicytes apoptosis was detected by the Tunel. DR5 expression was detected by the immunohistochemistry assay. ResultsCompared with the blank control group, the CIA model group showed a higher rate of ankle swelling, which showed that the model was successfully created. Compared with the model group, the ankle joint swelling rate was lower,joint damage was lighter, synovial cell apoptosis was more obvious and DR5 expression was higher in the hTRAIL-treated model groups, especially the 2μg hTRAIL CIA group, and in a dose-dependent manner.The difference was statistically significant(P<0.05). ConclusionshTRAIL enhanced synovicytes apoptosis, reduced the joint swelling and increased the expression level of DR5 in rats with collagen induced arthritis.
