Objective: The cognitive dysfunction, negative symptoms and positive symptoms were three core symptoms in schizopehrenia patients. 100first-episode schizophrenia patients were recruited as the subjects to explore the association between cognitive impairment and GABRB2 and PRODH genes,and to further explore the genetic mechanism of cognitive dysfunction in schizophrenia patients.Methods: Subjects: 100 patients with schizophrenia and 90 healthy volunteers were recruited, the volunteers were healthy Chinese han residents.Procedures: The cognitive dysfunction were assessed with the WSCT, ANT,Nback, TMT tests within one week after admission. All the subjects were extracted DNA from blood samples. 4 SNPs(rs383964, rs372055, rs450046,rs385440) in PRODH and 3 SNPs(rs194072, rs252944 and rs187269) in GABRB2 were selected to genotyped by sequencing techniques. Result analysis: The SPSS20.0 was used to analysis the data. The comparison of different cognitive indicators were analyzed by t test. And the genotype and allele of different SNPs were analyzed by chi-square, It indicated that significant differences were exited when the P was lower than 0.05.Results:1. Hardy-Weinberg equilibrium test: The genotype frequencies of the 7SNPs were not deviated from the Hardy-weinberg equilibrium(P>0.05) in allsubjects, illustrating that the samples come from natural population of random mating.2. Association between PRODH and schizophrenia: there was only one SNPs( rs385440) whose genotype and allele frequency had a significant association to schizophrenia(P=0.014,0.004).3.Association between GABRB2 and schizophrenia: we found no association between schizophrenia and the three SNPs of GABRB2.4. Comparison of the wrong number of AMTA, Nonperseverative Errors(NPE) of WSCT of patients with different RS383964 genotype showed significant differences(P=0.011,0.049).5. Comparison of the wrong number of TMT A of patients with different RS194072 genotype showed significant differences(P=0.049),and comparison of AA and AG genotype showed significant differences(P=0.025).6. Comparison of the Nface0 backRT with different RS194072 genotype shows significant differences(P=0.021), further analysis showed significance of CC,CG genotypes(P=0.008).Conclusions:1.The polymorphism of rs385440 in PRODH gene may be associated with schizophrenia in population of north of China. The proportion of AA,AG genotypes and allele A obviously increase.2.The cognitive function of patients with different genotypes in rs383964,rs450046, rs194072 SNPs exists obvious differences.3.Different Rs383964, rs450046 genotypes of PRODH and different rs194072 genotypes of GABRB2 may be the genetic mechanism of the cognitive dysfunction in schizophrenia patients.
