| Objective:To investigate the different expression of serum miRNAs between the preoperative malignant and the benign of ovarian tumor, to analyze its correlation with clinicopathological progress and prognosis features of epithelial ovarian cancer.Methods:â‘ Select 48 serum miRNAs which have been to be related to ovarian cancer and order custom miRNA cards. â‘¡Detect the expression level of the 48 serum miRNAs by TaqMan low density array in maligant and benign epithelial ovarian cancer. â‘¢Further confirm the candidate differentially expressed miRNAs in large samples using RT-PCR and analyse the relationship between the expression level of selected miRNA and clinical patheological factors, progress and prognosis.Results:â‘ TaqMan low density array showed that miR-20aã€miR-125b and miR-126 were significant different in the selected 48 serum miRNAs between the 18 malignant samples and the 16 benign samples(P<0.05). â‘¡Further RT-PCR showed that only miR-125b was significant different between 20 malignant serum samples and 18 benign serum samples (P<0.05). â‘¢miR-125b was significantly increased in 135 ovarian cancer patients as compared with 40 benign individuals. The expression of miR-125b was higher in early stage patients than advanced stage patients(P<0.05). The patients without residual tumor expressed miR-125b higher than patients with residual tumor. â‘£The high level of miR-125b was significantly correlated with longer progress free survival(PFS) (P<0.05), but it did not correlate with overall survival(OS) (P>0.05).Conclusion:miR-125b may play an important role in the pathogenesis of epithelial ovarian cancer and prognosis. It may a potential gene to predict the recurrence of epithelial ovarian cancer. |
