Objective:By detecting the impact of triptolide(tripterygium extract) on the biological characteristics of melanocytes, including melanocyte proliferation, melanin synthesis and tyrosinase activity. Understand the affect of triptolide on melanin synthesis, seeking new and secure way to treat hyperpigmentation disorders, provide more choice over for clinicians and patients.Methods:Use rat immortalized melanocytes(B10BR); The triptolide concentration set 1× 10-3,10-4,10-5,10-6,10-7 μg/ml 5 groups, and established the control group. To investigate the effects of triptolide on melanocytes proliferation, melanin synthesis and tyrosinase activity, melanocyte proliferation was measured by MTT assay, melanocytes melanin synthesis was measured by NaOH lysis method, tyrosinase activity was measured by DOPA oxidation.Conclusions and Significance:Melanocytes are very sensitive to triptolide, after single-factor analysis of variance:10-7μg/ml triptolide had no significant effect on melanocyte proliferation and tyrosinase activity, compared with the blank control group, P>0.05, there was no significant defference statistically; 10-5μg/ml and 10-6μg/ml triptolide had no significant effect on melanocyte proliferation, compared with the blank control group, P>0.05, but the tyrosinase activity were reduced, P> 0.05; 10-6μg/ml and 10-7μg/ml triptolide group had no significant effect on melanin synthesis, P> 0.05; 10-5μg/ml triptolide group of melanin synthesis has a certain extent, compared with the blank control group, P= 0.029<0.05; High concentration (10-3μg/ml and 10-4μg/ml) of triptolide on the proliferation of melanocytes, tyrosinase activity and melanin synthesis were significantly reduced, and the concentration was positively correlated, the difference was statistically significant. In conclusion,10-5μg/ml triptolide group can reduce melanin synthesis by inhibit tyrosinase activity,10-3μg/ml and 10-μg/ml triptolide groups can reduce melanin synthesis by inhibit tyrosinase activity and melanocyte proliferation. The results show that triptolide can inhibit melanocyte proliferation and reduce the activity of tyrosinase, reduce melanin synthesis, decrease pigmentation, provide a theoretical basis for triptolide as potential depigmentation agents.
