Objective:To probe into the function of immuno-regulation and inflammantory inhibition on New Yanghe Decoction by examining the level of interleukin-6 (IL-6), interleukin-17 (IL-17), interleukin-21 (IL-21) in serum and the expression of phosphorylated signal transducer and activator of transcription 3 (pSTAT3) in joint tissue, and to tentatively analyze its mechanism from cellular and molecular level.Method:Randomly divided 48 BALB/c mice into 6 groups including the normal group, model group, positive drug group, and low-dosage, medium-dosage, and high-dosage groups of New Yanghe Decoction. Mice were initially modeled by adopting the method of combining proteoglycan with Freund’s adjuvants intraperitoneally, and secondly modeled for enhancing after one week. Another one week later after modeling, mice in groups with medication started the intragastric administration once daily. Mice were executed by exsanguination until the 91st day after gagave started, and the spinal column and one side of the knee joints were dissected and collected. The concentration of IL-6, IL-17, and IL-21 in serum was inspected, the level of pSTAT3 in knee joint and spine column was examined by immunohistochemistry, and the pathological changes of the spine and the knee joint was observed.Result:The content of IL-6, IL-17, IL-21, and pSTAT3 in the model group were increased than those in the normal group(P<0.05). After medicine administration, the content of IL-6, IL-17, IL-21, and pSTAT3 in each group treated by Traditional Chinese Medicine were decreased than those in the model group (P<0.05 or P<0.01)Conclusion:1.The expression of IL-6, IL-17, IL-21, and pSTAT3 in model group was higher than that in normal group. The inflammatory infiltration on joint pathology indicates that cytokines such as IL-6, IL-17, IL-21, and pSTAT3 participate in the occurrence of ankylosing spondylitis(AS).2.New Yanghe Decoction can reduce the content of IL-6, IL-17, and IL-21, and inhibit the expression of pSTAT3, demonstrating that New Yanghe Decoction might treat AS by inhibiting the expression of IL-6, IL-17, IL-21, and pSTAT3.
