Objective:Glehniae Radix is the dry root of Glehnia littoralis Fr.Schmidt ex Miq.The Mongolian medicinal name is Chagan-sorlo,which is a commonly used Chinese and Mongolian medicine in clinic.It has been used for the treatment of respiratory and gastrointestinal disorders.Through Market Research and development,it is found that the processing methods of Radix Glehniae are not uniform in actual production,while the traditional peeling processing methods are complicated,time-consuming and laborious.At present,there are few studies on the chemical constituents of the root bark of Glehnia littoralis at home and abroad.Therefore,this study systematically studied the chemical constituents of the root bark of Glehnia littoralis.The quality standard of Glehniae Radix contained in the Chinese Pharmacopoeia?2015 edition?contains only two contents:character and microscopic identification,lacking the content determination method of effective ingredients,which can not systematically and completely reflect the quality of Glehniae Radix.Therefore,the fingerprint of Glehniae Radix was studied,and the contents of five main components were determined.The determination methods of total coumarin,total lignans and total phenolic acid were established.The aim was to provide scientific basis for rational utilization and quality evaluation of Glehniae Radix.Methods:The chemical constituents of ethyl acetate extract and n-butanol extract from 70%ethanol extract of root bark of Glehnia littoralis were studied.The structures of the monomer compounds were identified by modern spectroscopy(UV,ESI-MS,1H-NMR,13C-NMR,etc.).The fingerprints of Glehniae Radix were established by optimizing the chromatographic and extraction conditions.And a method for simultaneous determination of five components was established.The contents of total coumarin,total lignans and total phenolic acid in Glehniae Radix were determined by UV for the first time,and compared with the root bark of Glehnia littoralis.Results:1.Twelve compounds were isolated from the ethyl acetate extract of 70%ethanol extract from the root bark of Glehnia littoralis.11 of them were identified as psoralen?1?,esculetin?2?,scopoletin?3?,imperatorin?4?,kaempferol?5?,quercetin-3-O-glucoside?6?,vanillic acid?7?,isovanillic acid?8?,and3-O-caffeoylquinic acid?9?,?-sitosterol?10?,daucosterol?11?and adenosine?12?were isolated from the extract of n-butanol.Compounds 5,6 and 8 were isolated from this plant for the first time,and other compounds are being identified.2.An HPLC fingerprint of Glehniae Radix was established.There were 28 common peaks in the fingerprint,and 13 of them were identified as adenosine,syringin,chlorogenic acid,esculetin,caffeic acid,scopoletin,rutin,salicylic acid,psoralen,xanthotoxin,imperatorin,isoimperatorin and?-sitosterol.3.To establish a method for simultaneous determination of chlorogenic acid,rutin,xanthotoxin,imperatorin and isoimperatorin in Glehniae Radix.On this basis,the differences of components and contents between Glehniae Radix and the root bark of Glehnia littoralis were compared.4.A method for the determination of total coumarins,lignans and phenolic acids in Glehniae Radix by UV was established for the first time.The contents of total coumarin,total lignans and total phenolic acids in different batches of Glehniae Radix were 1.182.81%,2.6114.03%and 1.585.06%.Conclusion:Twelve monomer compounds were isolated and identified from the root bark of Glehnia littoralis,of which compound 5,6,8 was isolated from the plant for the first time.The established HPLC fingerprint of Glehniae Radix is typical,comprehensive and representative,which meets the technical requirements of fingerprint.The established method for the determination of total coumarin,lignans and phenolic acids in Glehniae Radix is simple to operate,with low equipment requirements and good reproducibility.It provides more basis and reference for the actual production and quality evaluation of Glehniae Radix.
