A novel enzymes assists-steam distillation system was developed for controlling Cinnamomum longepaniculatum(C.longepaniculatum) essential oils. The impact of various factors on the preparation of the C.longepaniculatum essential oils was examined. Based on single factor experiment, the response surface methodology was used to optimize the C.longepaniculatum essential oils preparation process. By using the optimal enzyme auxiliary conditions C.longepaniculatum essential oils was prepared that the dynamic model analysis steam distillation extraction of essential oil.Optimal conditions was used to extract the essential oil was studied by means of in vitro and in vivo antibacterial activity. The main contents were as follows:1. Through the single-factor enzyme extract C.longepaniculatum essential oils subsidiary conditions experimental study to determine the pH value, reaction time, reaction temperature influence on oil yield. And the extraction process of the adoption of various factors on the basis of single factor experiments to optimize the response surface to index by the yield of C.longepaniculatum essential oils, the best extraction condition:pH value of 5, reaction time 67min, reaction temperature is 48.5 ℃. Repeat 5 times under the condition of the verification experiment, yield of C.longepaniculatum essential oil was 4.37%, that was 1.5 times than that without the enzyme pretreatment group. And the experimental results verify the regression equation predicted values fit better, extraction conditions reliable.2. With C.longepaniculatum essential oils for the study, based on dynamics model of Vt= V∞(1-e-kt)by enzymes assisted-steam distillation of extraction of C.longepaniculatum oils and experiments validate the dynamics model. Fitting dynamics model for extracted process by enzymes assisted-steam distillation of C.longepaniculatum essential oils, with the parameters obtained to calculate the amount of essential oil extracted with the experimental values coincide dynamics model can be described enzymes assisted-steam distillation of extraction process. By GC/MS detection and analysis, distillation time was 120min excellent quality of essential oil of camphor oil, enzymatic assisted-steam distillation of the best distillation time 120min.3. C.longepaniculatum essential oils for the 15 kinds of tested strain have a certain inhibitory effect, which has a broad-spectrum antimicrobial properties. Measured the oils for MIC and MBC, Where the best of Staphylococcus aureus and yeasts. And in 2.0,1.0,0.5 μL/ mL concentration of essential oils, as the culture time increased, the effect of oil camphor oil Staphylococcus aureus and Escherichia coli best.4. A preliminary study of the in vivo antibacterial activity of C.longepaniculatum essential oils in mice, the first after several virulence tests, the mortality rate of mice was maintained at 80%-100% of the diluted bacterial concentration. The bacteria-infected mice with different doses of essential oils on mice fed, compare the following items:the number of dead mice, survival time after infection, and the number of colonies in their blood by after 6h, the results showed that the C.longepaniculatum essential oils of infected mice has a protective effect, indicating that it has some antibacterial activity in vivo.
