Thymoma is the most common anterior mediastinal primary tumor, originated in the thymic epithelial cells. In adults, the incidence rate is about 20%, 50% in anterior mediastinal tumor, of the total population incidence rate is about 0.0015‰.Thymoma is a rare and unique tumor with a long natural history that makes it difficult to study. Consequently, there is a dearth of prospective diagnostic or therapeutic clinical trials. To our knowledge there has not been an analysis of conditional survival of thymoma in the literature. In order to provide a specific biological marker for the diagnosis of thymoma, open up new therapeutic approach.In recent years, more and more researchers focus on research the onset, progression and metastasis mechanisms of thymoma.In early experiments, we had measured cell morphology, cell growth curve and doubling time of testing, tumorigenicity situation observed, expression of immunocytochemistry associated antigen for mature thymoma cell lines Thy0517. In the current experiment, we will conduct a surface antigen expression, ultra structural of the electron microscope and genetic characteristics(chromosome aberrations)analysis of thymoma cell lines Thy0517. Provide a better basis and experimental material to study the incidence, development and transfer mechanisms of thymoma.Objective:Identified surface antigen expression, ultra structural electron microscope observation and genetic characteristics(chromosome aberrations) on established human cell lines thymoma cells Thy0517. Provide good experimental materials to study the incidence progression, metastasis mechanisms and explore new clinical approach of thymoma.Methods:Slow freezing and rapid thawing was used to cryopreserve and recovery the established a thymoma cell lines Thy0517, tryptic digestion technique was used to passage cells, phase contrast microscope to observe cell morphology, flow cytometry to cell surface antigen expression of CD9, CD29, CD54, CD58, Giesmsa banding techniques to observed cytogenetic change, Transmission electron microscope(TEM)to observed the cell ultrastructure of thymoma.Results:Cell morphology and growth cycle stability, cells were observed by 200 times phase contrast microscope. Compared with the 10 th generation and 40 th generation cells, the cells were no significant change in morphology. Flow cytometry showed that, compared with the negative control group, CD9, CD29, CD54, CD58 expression was positive in thymoma cell surface Thy0517. G-banding analysis showed that,thymoma cell lines Thy0517 contain some common chromosomal aberrations of type AB thymoma, and exhibits a number of other chromosomal aberrations its unique.TEM observed cytopathic follows that, cell morphology and cell nuclei intact, nuclear shape irregular, the proportion of nuclear and plasma increases, nuclei within rich heterochromatin, nucleolus big and close to the edge, two or more visible nucleoli,these are the characteristics of malignant cells. Intercellular connections are mostly generic, desmosomes can be seen, and these reflect the structural characteristics of epithelial tumors.Conclusion:Human thymoma cell lines Thy0517 cell morphology and growth cycle stability,Able to express common surface antigen of some thymoma cells, exist common chromosomal aberrations of type AB thymoma, have the characteristics of tumor cells in ultrastructural. These confirm that thymoma cell line Thy0517 is a stable cell line which has the features of AB thymoma, and can provide a good experimental material to study the incidence, development mechanisms and new therapeutic approaches in clinical of thymoma.
