Objective : To develope a stable animal modle of minimal persistent inflammation in allergic rhinitis mice, have a preliminary understanding of MPI and provide an experimental basis for further research on the MPI period.Methods: Sixty female SPF BALB/c mice were randomly divided into four groups, each groupe has fifteen animals.Group A is positive control group, group B as experimental group(MPI group), group C negative control group and group D for blank control group.Group A, B, C received intraperitoneal injection weekly for 3 weeks with a PBS solution containing OVA mixed with AL(OH)3. From day 22,Group A, B, C were challenged intranasal daily with 40mg/ml OVA with one week. After the successful establishment of the AR model, from day 29, group A, B, C mice were challenged with 40mg/ml OVA,4mg/ml OVA and PBS respectively for a long-term intranasal stimulation,lasting for three weeks, groupe D were always conducted with PBS solution instead of OVA for sensitization and stimulation.Then the mice were assesed by nasal symptom, the degree of nasal mucosal eosinophil infiltration, expression of ICAM-1 in the epithelial cell.Results:After nasal stimulation, AR models were successfully established in groupe A, B,and C, the symptomatology score had no significant difference among the three groups of mice(P>0.05); When challenged with 4mg/ml OVA solution intranasally, the nasal symptom such as sneezing, scratching and other symptoms gradually disappeared in MPI group(group B),he difference compared with group A had statistical significance(P <0.01),there was no significant difference compared with group C and D(P>0.05), but therewas still some eosinophil infiltration in nasal muosa and mild ICAM-1 expression on the epithelial cells, and the differences compared with group C and D had statistical significance(P <0.01).Conclusions: By sensitized and challenged with OVA, BALB/c mice can be successfully established allergic rhinitis model; On the basis of establishing stable AR model in mice,we can develop the minimal persistent inflammation models in allergic rhinitis mice by giving low concentration OVA nasal stimulation for a long time. This method has advantages of simple operation, high success rate, good repeatability, providing an experimental basis for further deep study and good treatment of MPI of allergic rhinitis.
