Gastric cancer(GC) is a major contributor to the global cancer burden, ran king as th-e fifth most commonly diagnosed cancer.Worldwidely,it is the third l eading cause of cancer death in male and the ?fth in female.Previously,Three g enome wide associatio-n stud-ies found phospholipase C epsilon 1(PLCE1)gene,which can encode PLCE1 p-rotein.this protien belong to the phospholipase fa mily.Founction as secondary messe-nges,inositol-1,4,5-trisphosphate and di acylglycerol,which formed following th-e hydr-olysis of polyphosphoinositides th at catalyzed by PLCE1 protein,subsequentl-y involve in cell growth and differe ntiation. New diagnostic basis of gastric cancer ma-y be fou-nd by in-depth st udy.Meanwhile,it can provide a theoretical basis for Scree-ning of h-igh-risk g roups,as well as the prevention of vulnerable groups. At present, t-he PLCE1 h a-s become a hotspot in the field of cancer research. Objective:All of this study is to investigate the relationship between PLCE1 rs2274223(A/G) of single nucleotide polymorphisms(SNP) and gastric cancer,which provides a series of genetics reference basis for screening, early diagnosis,and treatment among high-risk population,otherwise,it also help setting individualized preventive measures. Methods:114 cases of the hui nationality we collected from Qinghai University Affiliated Hospital as the case group during August 2012 to August 2015.Meanwhile,other 157 healthy controls from the medical center were matched by age and sex in our Case-control study. Sequenom Mass ARRAY system was utilized to detect the genotype of rs2274223 and analyze the distribution of of the difference of rs2274223 in these groups.SPSS19.0 was used for all data statistics processing,inspection comparison between two groups about their basic situation, including gender, age, family history of cancer as well as smoking and alcohol status, chi-square test and Hardy- Weinberg balance analysis were used to test the genotypes and alleles of rs2274223.By binary Logistic regression method to calculate the ratio of the relative risk and 95% confidence interval. Results:1)Rs2274223(A/G) allele including AA、AG and GG three points. 2)In gastric cancer group,rs2274223 PLCE1 gene locus on AA, AG, GG and AG +3) GG genotype frequency were 46.5%, 47.1%, 6.4% and 53.5%. The corresponding frequency in the normal control group was 57.0%, 33.3%, 9.7% and 43.0%.Compared in two groups,There were no statistically significant difference between AG、GG and AA(p>0.05);Compared with AA, including the mutation point of all genotypes AG + GG distribution no differences between two groups(p>0.05);Comparison between alleles A and G,the differences still have no statistically significant difference between the two groups(p>0.05). Conclusion:Rs2274223 PLCEl gene has no obvious correlation with the incidence of region of qinghai hui gastric cancer.
