| Objective:To observe the expression of AMPK, the transshipment of GLUT4 and the ex pression of VAMP isoforms, including VAMP1ã€VAMP2ã€VAMP3ã€VAMP4ã€V AMP5ã€VAMP7ã€VAMP8, in the 3T3-L1 adipocytes stimulated by different doses of Metformin.Methods:1.3T3-L1 fibroblasts of mouse were cultured in the environment of 37 ℃ and 5%carbon dioxide, differentiated into mature adipocytes by inducing conditional media and identified by red oil O.2.Detect the expression of GLUT4,VAMP1,VAMP2,VAMP3,VAMP4,VAMP5,VAMP7 and VAMP8 with RT-PCR after the stimulation of different concentration of Metfo rmin.3.Analyze the statistical data using SPSS13.0 software and record MD±SD as dat a representation. The differences among multiple groups were compared with randomize d analysis of variance and the comparison among groups were performed with LSD test.P < 0.05 for the difference was statistically significant.Results:1.3T3-L1 fibroblasts of mouse were successfully differentiated into mature adipocyte s by inducing conditional liquid.2.The results of RT-PCR:1)The results showed that compared with the control group, the expression of AMPK and GLUT4 were statistically difference(P<0.05), sim ultaneously, Metformin stimulated AMPK expression and GLUT4 translocation in a dose-dependent manner. 2)The expression of VAMP2,VAMP3 and VAMP8 were sta tistically Significant compared with the control group(P<0.05), and the expression of these VAMP isoforms were dose related with the concentration of Metformin.However, the expression of VAMP1,VAMP4,VAMP5 and VAMP7 had no statistical difference with the control group(P>0.05).Conclusion:Metformin up-regulated the expression of VAMP2, VAMP3 and VAMP8 in a dose-dependent manner. However, the regulation of Metformin in the expression VA MP1,VAMP4,VAMP5 and VAMP7 needs further study and exploration.The results s uggested that Metformin can up-regulate the expression of some of VAMP isoforms,which plays an important role in the trafficking of GLUT4 in adipocytes. |
