Objective:To explore the relationship among the expression of CEACAM1 in glioma patients,the dysfunction of T cell and disease progression, we studied the expression of CEACAM1 in CD4+ T cell, CD8+ T cell of peripheral blood and CD8+ T cell of tumor infiltrating lymphocytes, and further discussed the correlation of surface inhibitory molecules between peripheral blood lymphocytes and TILs.Methods:1. We marked with antibodies on T cells in peripheral blood to identify CD3,CD4,CD8 and CEACAM1, and detected the expression of CEACAM1 on the surface of CD4+ T cell and CD8+ T cell by flow cytometry. Then we separated tumor infiltrating lymphocytes in tumor tissue and detected the expression of CEACAM1 in the same way as before.2. We quantitatively analyzed the secretion of TNF-α in peripheral blood of glioma patients.3. Patients’ clinical data, karnofsky performance status(KPS) and degree of functional disability were collected and calculated.Results:1. After detecting the expression of CEACAM1 on peripheral blood T cells of patients and healthy volunteers, we found that glioma patients had a higher level of CEACAM1 on CD4+ T cell and CD8+ T cell of peripheral blood than healthy volunteers.2. The expression of CEACAM1 on CD8+ T cell of tumor tissue is statistically higherthan that of normal brain tissue.3. In peripheral blood, patients with high-grade giloma had a remarkably higher levelof CEACAM1+CD8+ T cell than those with low-grade giloma. The level ofCEACAM1+CD4+ T cell was also higher in patients with high-grade glioma, but nostatistical significance was found.4. In tumor infiltrating lymphocytes, the level of CEACAM1+ CD8+ T cell was higher in patients with high-grade glioma than those with low-grade glioma.5. Patients with glioma, especially those with high-grade glioma, had a remarkably higher level of CEACAM1+CD8+T cell in tumor tissue than that of peripheral blood.6.We found a negative correlation between CEACAM1+CD8+ T cell level and KPS,however, no significant correlation was found between CEACAM1 +CD4+ T cell level and KPS.7. We found a negative correlation between serum TNF-α level and peripheral CEACAM1 +CD4+ T cell and CEACAM1 +CD8+ T cell level.Conclusion:Level of CEACAM1+CD4+T cell and CEACAM1+CD8 +T cell was up-regulated in peripheral blood of glioma patients, and it was even more remarkable in high-grade glioma patients. There was a negative correlation between CEACAM1+CD8+T cell level and KPS,as well as between serum TNF-α level and peripheral CEACAM1+CD4+ T cell and CEACAM1 +CD8+ T cell level. Our present study indicated that CEACAM1 maybe an important marker in glioma progression, and further more, maybe a potential target of giloma treatment.
