| This thesis aimed to preparing floatable granules of traditional Chinese medicine.It was according to a certain method to extracting and concentrating the traditional Chinese medicine, the compound liquid medicine was coating to the core of floating extruded feed pellet. And then the core was coating another liquid drug formulation. The coated drug was delaying to fully release abount 10~15 minutes in water. Relative to the general terms of drug particles for fish, it can reduce drug dissolving in water before fish eat it. It was suitable for fish feeding habits, and it was used to preventing and treating the bacterial diseases,viral diseases of fish. Compared with traditional fish medication, granules was used in traditional Chinese medicine to solving the problem of excessive. It was ensuring fish nutrition while treating fish diseases.According to the physical and chemical properties of the main component in the prescription medicines, the detection index was an active ingredient content. This thesis was established a measurement method of index components of epigoitrin, icariin and the Astragaloside by High Performance Liquid. It was using the method of single factor analysis and orthogonal analysis, to studying and optimizing the extraction mothod of Radix, Astragalus and epimedium. The results showed that the best extraction of radix isatidis was as follows: it was immersing in water 60 minutes with 15 times the ratio of liquid, and then it was extracted with water 105 minutes 2 times; It was showed that the best extraction of epimedii folium was as follows: it was immersing in water 90 minutes with 30 times the ratio of liquid, and then it was extracted with water 75 minutes 3 times. It was showed that the best extraction of astragalus membranaceus was as follows: it was immersing in water 90 minutes with 25 times the ratio of liquid, and then it was extracted with water 105 minutes 3 times. After extracting,it was filted several times and then condensed the liquid into each 1m L containing crude drug 0.5 g complex concentrates herbs bya rotary evaporator at particular conditions. Last it was mixed of the mass ratio of Radix, Astragalus and Epimedium with 2: 2: 1.After extraction and concentration, it was explored the coating conditions. It was coating with two layers of the granules, first concentrated liquid encapsulated in the particle surface, and then wrapped in a controlled release of the sustained-release layer.And then it was examining the mothod of preparation floatabe Fuzhengjiedu granules accoding to determining the rate of drug release and examining the coating liquid and the incresae weight of liquid coating.The results showed that, The best method to preparing floatable Fuzheng Jiedu Granules was: after coating a 7.23% of increaing weight of the concentrated liquid, it was coated with a 5% increaing weight of 7.76% of the solids content solution of ethyl cellulose; and then the granules were drying in the oven 2h at 55 ℃. The epigoitrin was as indicators to determin the content of drug by HPLC, the drug was released less than 30% in 3minute, and release more than 80% in 15 min in the water.The granules were identified by thin layer chromatography. Accoring to studying it’s stability by influencing factor test and accelerated test, the experimental results that the stability of the product was good, the test results was conforming the requirements of the pharmacopoeia.In summary, floatable fuzhengjiedu granules’ s extraction and preparation process was reasonably. The quality of products can be effectively control, and the stability of formulation was good. It was achieved the desired objectives, and it can be used for preventing and treating the bacterial diseases and viral diseases of fish with practical significance. |
