ObjectiveTo explore the possible mechanism and therapeutic target of Hedysarum Polybotys saccharide to Diabetic Rats induced by streptozotocin by observing the expression of NF-κB,TNF-α,IL-1β and the effect of retinal histopathological changes.MethodsThere are 80 male wistar rats, randomly divided 10 rats as the blank group, the remaining 70 was made diabetic rat model by continuous intraperitoneal injection for3 days of streptozotocin(streptozotocin,STZ,30 mmol.kg-1). A week later,60 qualified model rats were randomly divided into 5 groups,12 rats in each group: All treatment groups were respectively with Calcium dobesilate group(90mg.kg-1.d-1),HPS(150,100 and 50mg.kg-1.d-1),the blank group and model group with equal volume of physiological saline by gavage once daily, 8-week period continuous.Then,we peel eye retinal to have pathological observation and to observe the expression of NF-κB and TNF-α by immunohistochemical staining.At the same time, we take the blood of rats to the content of IL-1β in serum by ELISA,The average absorbance value of positive cells was calculated by Image-Pro Plus image analysis software. The experimental data were analyzed by using SPSS19.0 statistical analysis software, and the method was based on single factor analysis of variance.Results1.In blank control group, each layer of the rats’ retina arranged orderly and have normal forms of organization structure and morphology without exception. In model group, rats’ retinal layers have different degrees of edema and sparse disordered structure,the decreased number of inner and outer nuclear layer cells,telangiectasia in nerve fiber layer and ganglion cell layer,and different degrees of thinningin each layer.Compared to the model group, the retinal pathological damage get different degrees of improvement in HPS high, medium and low dose group. In HPS high dose group,each layer arranged almost normal, no obvious edema, retinal thickening.2.Compared with the blank group, the IL-1β expression in the serum of DM model was significantly higher(P<0.01);Compared with the model group,levels of IL-1β in the serum of the treatment groups were decreased(P<0.05), and the HPS high dose group was the most significant(P<0.01).3.Compared with the blank group, the expression of NF-κB,TNF-α in rats’ retina of model group and the treatment group were increased(P<0.05, P<0.01);Compared with the model group,the treatment groups were decreased,HPS high dose group has significant difference(P<0.05, P<0.01),HPS high dose group was the most significant(P<0.01);With the increase of the dose of HPS in the treatment of gastric lavag, the area of brown particles in the retinal tissue was gradually decreased.Conclusion:1.HSP can significantly improve early damage of the retinal morphology in the DM model rats induced by STZ, which indicates that HSP can obviously protect theretinal tissue of DM model rats.2.HPS can significantly decrease the content of IL-1β in plasma of DM model rats,and reduces the degree of inflammation in DM rats.3.HPS can repress the retinal neovascularization by regulating the expression of NF-κB,TNF-α in retinal tissue of diabetic model rats,.DR lesions may be blocked by inhibiting the expression of NF-κB,TNF-α.
