The research in this paper is focused on two aspects:Pharmacokintics research of Hedysarum polybotys saccharide 3?HPS3?and Pharmaceutical research of Jiexin Huatan Tongqiao Cataplasm.HPS is one of the most important components of Radix hedysari.At present,there are many reports on the structure,activity of HPS and little on its molecular mechanism and metabolic kinetics.Based on the fact that HPS3 is the most active component of HPS,the distribution and metabolism of HPS3 in mice were studied in order to provide a theoretical basis for the development and application of HPS.As a modified preparation,Jiexin Huatan Tongqiao Cataplasm?JHTC?is derived from the traditional classicsal prescription,consists of Semen brassicae,Rhizoma corydalis,Asarum,Euphorbia kansui,fresh ginger.In this paper,the extraction process,the preparation process and the quality standard are studied.The concrete research contents are as follows:The first part:Pharmacokintics research of HPS3?1?FITC fluorescence labeling of HPS3 and the distribution and Metabolism of HPS3-FITC in miceFluorescein isothiocyanate?FITC?as fluorescence probe was linked up with HPS3,and the fluorescence degree of FITC was 1.71%.Spectrophotofluorimetry method was established to study the distribution and metabolism of HPS3-FITC in mice.The results showed that the half-life t1/2/2 of HPS3-FITC was 5.550 h.HPS3-FITC was mainly distributed in kidney and liver tissue,with a significant kidney and liver targeting effect.?2?ANTS fluorescence labeling of HPS3 and the distribution and Metabolism of HPS3-ANTS in mice8-aminonaphthalene-1,3,6-trisulfonic acid disodium salt?ANTS?as fluorescence probe was linked up with HPS3.the fluorescence degree of ANTS was 2.97%.High-performance gel-permeation chromatographywas established to study the distribution and metabolism of HPS3-ANTS in mice.The results showed that the half-life t1/2/2 of HPS3-ANTS was 4.294 h.HPS3-ANTS was mainly distributed in kidney and liver tissue,and metabolized by liver and kidney tissue.The second part:Pharmaceutical research of Jiexin Huatan Tongqiao Cataplasm.?1?The extraction process of JHTCThrough single factor experiment,the best extraction time of Asarum volatile oil by Steam extraction method was 8 h.The yield of dry paste and the content of tetrahydropalmatine were used as the evaluation index to design the orthogonal experiment.The optimum process was that:Extraction solvent was 70%ethanol;the ratio of material to 70%ethanol was 4:1 mL/g;the time of extraction was 1h and Extraction times was 3.?2?The preparation process of JHTCOn the basis of determining the amount of calcium chloride,azone and drug extract powder,the orthogonal design of the dosage of sodium polyacrylate,gelatin and glycerol was carried out according to the comprehensive score of sensory index and physical character.the best matrix prescription for JHTC as follows:sodium polyacrylate 1.5 g,gelatin 1.5 g,glycerol 6 g,azone 0.8g.calcium chloride 0.04 g.?3?The quality standard of JHTCSemen brassicae,Rhizoma corydalis,Asarum,Euphorbia kansui of JHTC were identified by TLC.The content of tetrahydropalmatine,Sinapine thiocyanate and asarinin were determined by HPLC.The limit of content is not less than 0.6996mg as Sinapine thiocyanate and not less than0.0552mg as asarinin in one piece of JHTC?0.25 g?.
