Objective In this research, we deteced the expression about CD4+CD25+Foxp3+Tregs(Regulatory T Cell) in peripheral blood, which contained CINⅠ CINⅢ, invasive cervical cancer patients(had surgery), invasive cervical cancer patients(had no surgery);and then we analyzed the reasons for the situation; last, we deeply investigated the role and significance in the cancer patients especially in invasive cervical cancer patients; The purpose of our study is that we will provide the basis for the immunological treatment of invasive cervical cancer. Methods 1.The object of our research were CINⅠ CINⅢ patients, invasive cervical cancer patients; and the invasive cervical cancer patients were divided into before surgery patients and after surgery patients by us; and we detected the change about the CD4 + CD25 + Foxp3 + Treg cells. We can also assess the immune status in the patients through the change. By the way, we explored the different immune status in the invasive cervical cancer patients; 2. Part Ⅰ: the groups of this study was divided into four groups by us, the 68 cases came from department of gynecology during 2014.32015.3 who treated in Gansu Provincial People’s Hospital, and the 29 cases was healthy female volunteers who treated in Medical Center. Then the group named: CINⅠ CINⅢ(22), invasive cervical cancer(had surgery,22), invasive cervical cancer( had no surgery,24), healthy people(controls,29); 3. Part Ⅱ: Collected the blood early in the morning and fasting; the blood of the reseach was needed 2ml for each patient, and placed in 4 ℃(Because the blood samples need to maintain freshness,the requirement of experiment blood samples collected within 48 h available to detect). All the samples needed fluorescent antibody staining, and then separated CD4+CD25+Foxp3+ Tregs by flow cytometry; last, we analyzed the data by Flow Jo7.6.1(The flow cytometry analysis software); 4. Part Ⅲ: t-test by SPSS17.0 was used for statistical analysis by, and then obtained the statistical results. Result 1.The expression of CD4+CD25+Foxp3+Tregs were presenting in each group; obviously,the degree of expression showed a significant difference; 2. The expression of CD4+CD25+Foxp3+Tregs were as follows: invasive cervical cancer(preoperative) group(4.012 ± 0.784)% was higher than CINⅠCINⅢ group(2.209 ± 1.228)%; invasive cervical cancer(preoperative)(4.012 ± 0.784)% was higher than invasive cervical cancer(after surgery) group(1.915 ± 0.508)%; invasive cervical cancer(preoperative) group(4.012 ± 0.784)% was higher than the healthy group(control group)(0.643 ± 0.375)%. And the data had statistically significant difference((P <0.05); 3. But when we analyzed the expression of CINⅠCINⅢ group, invasive cervical cancer(after surgery) group, and the healthy people(control group), we found that they had no statistically significant(P>0.05). Conclusion and significance In this research, we found that the expression of CD4+CD25+Foxp3+Tregs in invasive cervical cancer patients higher than the other groups; therefore we speculated that CD4+CD25+Foxp3+Tregs had different mechanisms to inhibit the immune effector cells, so that cancer cells produced immune escape. And CD4+CD25+Foxp3+Tregs cells are involved in the occurrence and development of invasive cervical cancer.
