Objective:To observe the osteogenic effects of recombinant human bone morphogenetic protein 2(rh BMP-2) combined with Bio-Oss to repair the defects of Beagle alveolar bone, and provide a better choice about the treatment of the regeneration of clinical alveolar bone defects.Methods: 1.Experimental animals and groups: 4 healthy 12 to 18-month-old male Beagle dogs were selected and divided into three groups according to the different section of the experimental teeth. Each beagle dogs the left maxillary, right upper and the left mandibular second, third premolar as experimental teeth, which the left maxillary as blank control group(control group): only basic treatment. The right maxillary bone group(Bio-Oss group): basic treatment + Bio-OSS + Bio-Gide. The left side of mandibular into experimental group(rh BMP-2 group): foundation treatment +Bio-Oss+ Bio-Gide+ rh BMP-2. 2.Experimental treatment: The U type of alveolar bone defects which was 5 mm depth and 3 mm width were prepared on the second and third premolar root. And the blank group did not do any treatment directly on the gingival flap suture. Bio-Oss group filled with Bio-Oss at the U type defects, then covered with Bio-Gide membrane for guided, suture. The rh BMP-2 group in the defect disposal to the marrow and filled with rh BMP-2/Bio-Oss and covered with Bio-Gide biological membrane for guided, then suture. 3.Imageological diagnosis: 2 Beagle dogs were sacrificed after operation for 8W and 12 W, the head was employed by spiral CT scanning, and the interception of jaw fragments were employed by Micro-CT scanning, observe the amount of alveolar bone regeneration in each group.4.Histological examination: Each experimental teeth from the original defect area was divided into proximal and distal parts, a part of conventional decalcified tissue sections stained with HE staining, and the other part of undecalcified for hard tissue slicing TB staining, qualitative observation of the operation, the new district of alveolar bone, cementum and periodontal ligament formation conditions and the basic structure.Results: 1.According to spiral CT scanning results, three groups were observed the formation of new bone after operation for 8W and 12 W. The rh BMP-2 group is the best, Bio-Oss group is the second,the blank group is the worst, the difference between the three groups have statistical significance(P < 0.05). 2.The results of Micro-CT scaning showed that three groups were observed the formation of new bone after operation for 8W and 12 W. Compared between BV, BV/TV and Tb. N, rh BMP-2 group is the best, compared with the other two groups have statistical significance(P < 0.05) 3.Tissue slicing HE staining showed that after 8W operation group can be observed the formation of new bone, that the new bone and cementum adhesion occurred in blank group, Bio-Oss group the new bone was more mature than the blank group, the rh BMP-2 group showed a large number of osteoblast rows are arranged in a row in the ossification center, become active in the process of ossification, the newly form ed periodontal ligament fiber are sat on new form alveolar bone; after operation for 12 W Bio-Oss group can be observed in the new form of periodontal membrane fiber bundles are embedded in the newly formed alveolar bone,rh BMP-2 group showed a newly formed cementum. 4.Hard tissue slicing TB staining showed that after 8W operation can be observed the formation of new bone, the blank group, only a small amount of new bone formation and calcification was not obvious, Bio-Oss group new bone formation compared with the blank group, bone trabecular bone permutation was disorder, rh BMP-2 group observe the bone repair materials is absorbed, which was surrounded by a newly formed bone, calcification was not obvious; after 12 W operation the bone were more mature than 8W, visible the calcifiedosteoin, Bio-Oss group shows the haversian system, rh BMP-2 group visible part of the new bone calcification, completely harvard and the system structure is clear.Conclusion: rh BMP-2 has a certain role in promote the regeneration of periodontal tissue defect, combination with Bio-Oss can obtain better effect of periodontal hard and soft tissue regeneration.
