Objective: To study the facet joint fusion with PLGA made of sheep lumbar facet joint bone fusion, using imaging technique、histology and molecular biology techniques for bone fusion was observed to provide a theoretical basis for the efficacy of bone fusion. Methods: 27 female goats(weight 30 ± 2Kg) randomly divided into experimental and control group, blank control group, each group of 9.Experimental group: removed goats bilateral L4-5 facet joint articular cartilage, then in the joint space built into degradable facet joint fusion; control group: removed goats bilateral L4-5 facet joint articular cartilage in the corresponding position with the implant fusion device of comparable size and autogenous block; blank control group: only removed the goats bilateral L4-5 facet joint articular cartilage, articular cartilage does not eliminate the lumbar facet joint placement of any implant. These sheeps were killed after 3、6、9 months respectively, to take stuffed whole lumbar spine, with the naked eye、X-ray、multislice spiral CT 、slicesHE staining and immunohistochemical Chemical Technology(OPG) of the specimens were observed. Results: Radiology:After 3 months,three groups showed no facet joint fusion, with the passage of time, the fusion rate of the three groups showed an increasing trend. After 6 months, the degree of integration of the lumbar facet joints in the experimental group was less than the control group and the control group, the experimental group compared with the control group, p=0.567, after comparison, the control group and the blank control group showed no statistical significance. After 9 months, the fusion rate of lumbar facet joints in the experimental group was better than the control group and the blank control group, after statistics, the experimental group compared wit the control group, p=0.0399, between the performance data shown significant differences. HE staining: After 3 months, the chondrocytes of the control group was most among three groups, the chondrocytes of experimental group was least compared with the control group, P < 0.01, there are significant differences; After 6 months, the experimental group had a number of chondrocytes, the number of chondrocytes in the control group had been significantly reduced, two sets of data, P<0.01, statistically significant; This time, the number of cartilage cells in the control group and the blank control group began gradually reduce, due to the chondrocytes transformed into bone cells,two sets of data, P>0.05, no significant difference. At 9 months, chondrocytes began to reduce in the experimental group, had completed most of the facet joint fusion, contrast with the control group P<0.01, statistically significant; blank control group were slightly in the number of cartilage cells was less than the control group for each time, no significant difference between two sets of data. Immunohistochemistry experiment:Before 6 months, the experimental group OPG expression in the previous were lower than the control group; at 3 months, two sets of data, P<0.01, statistically significant, at 6 months, the two groups data was not statistically significant; at 9 months, the experimental group was significantly higher than OPG, two sets of data, P<0.01, statistically significant, indicating that OPG obtain sustained expression in the experimental group, and was not because fusion degradation products to reduce the expression of OPG. Control group compared with the blank control group, the control group OPG optical density values were more than the blank control group, at 3 and 9months, the two sets of data, P>0.05, not statistically significant, in 6 months, two group data comparison P<0.01, statistically significant. At 9 months, among CT prompted unfused facet joint specimens, this sample optical density at different time points OPG average values were less than the overall average value, and pairwise disjoint data was statistically significant, indicating that the low expression of OPG, would directly affect the small joints of bone fusion. Conclusion: 1. Biodegradable lumbar facet joint fusion may provide initial stability of the lumbar facet joints of the lumbar facet joints can promote bone fusion, thereby enhancing fusion lumbar facet joints. 2. Lumbar facet joint fusion can be completely degraded in vivo expression of a fusion of the lumbar facet joint bone OPG does not adversely affect the degradation process. 3. Lumbar facet joint fusion of use, you can maintain the expression of OPG bone fusion zone late on facet joint fusion have a positive effect.
