Objective:To research the effect of compatibility of Salvianolic acid B, C on the expression of CCL2 and CCL3 in HK-2 induced by human serum albumin. Methods:Human renal tubular cells were randomly divided into six groups(the control group, the compatibility group, antibody group, salvianolic acid B group, salvianolic acid C group and salvianolic acid B and C group). In addition to the normal group, the other groups were given HSA induction. after the synchronization of 24 h.The treatment groups were given corresponding drug culture, cells were collected in 24 h,48h and 72 h respectively. The expression of PDGF-C, PDGFR-α, CCL2 and CCL3 were detected by Western Blot, Immunocytochemistry and Immunofluorescence. The levels of SOD, GSH and MDA were determined by WST-1 method, DTNB method and TBA method respectively. Results: 1. The results of PDGF-C 1) PDGF-C expression signal were brown expression sites mainly localized in the cytoplasm of HK-2. 2) Compared with the model group, the level of PDGF-C in the treatment groups were significantly decreased in 24 h, 48 h and 72h(P < 0.05). 2. The results of PDGFR-α 1) PDGFR-α expression signal were brown expression sites mainly localized in the cytoplasm of HK-2. 2) Compared with the model group, the level of PDGFR-α in the treatment groups were significantly decreased in 24 h, 48 h and 72h(P < 0.05).At the point of 72 h, the expression of PDGFR-α in the compatibility group and the antibody group was significantly decreased than the SAB group and the SAC group(P < 0.05). 3.The results of CCL2 1) The positive expression of CCL2 was green, the expression was mainly localized in the cytoplasm of HK-2. 2) Compared with the model group, the level of CCL2 in the treatment group was significantly decreased in 24 h, 48 h and 72h(P < 0.05). At the point of 72 h, the expression of CCL2 in the compatibility group and the antibody group was significantly decreased than the SAC group(P < 0.05). 4.The results of CCL3 1) The positive expression of CCL3 was green, and the expression was mainly localized in the cytoplasm of HK-2. 2) Compared with the model group, the level of CCL3 in the treatment groups were significantly decreased in 24 h, 48 h and 72h(P < 0.05). 5. Results of SOD, GSH and MDA: Compared with the normal group in 24, 48 and 72 hours, the level of SOD and GSH were significantly lower(P <0.05) and the level of MDA was significantly higher(P <0.05).Compared with the model group, the level of SOD and GSH in the treatment groups were significantly higher in 24 h,48h and 72h(P <0.05) and the salvianolic acid B+C group was significantly higher than the SAB group and the SAC group(P <0.05). Compared with the model group in 24, 48 and 72 hour, level of MDA in the treatment groups were significantly lower(P <0.05) and the salvianolic acid B+C group was significantly lower than the SAB group and the SAC group(P <0.05). Conclusion: Salvianolic acid B, C and drug compatibility could reduce the expression of PDGF-C, PDGFR-α, CCL2 and CCL3 protein, effectively enhance the activity of SOD, GSH, and reduce the amount of MDA. It suggests that the salvianolic acid B, C and drug compatibility pass through intervention of the PDGF-C/PDGFR signal pathway, reduce the inflammatory reaction and relieve oxidation reaction, which might play a role in delaying the process of RIF.
