Objective The goal of this study was to examine the relationship between OAS1(rs1131454)gene polymorphism and severity of Enterovirus 71(EV71) infection. We can explore the effects of different genotypes on risk of EV71 infection.Methods This study was performed in 216 patients with EV71 infection and 441 healthy control subjects. Genomic DNA of white blood cell were extracted and clinical characteristics of the case patients were collected. EV71 infection was diagonosed by a reverse-transcriptase polymerase chain reation(RT-PCR). The A/G SNP in exon3(rs1131454) of OAS1 gene was studied using polymerase chain reaction followed by restriction fragment length polymorphism(PCR-RFLP) method.Results 1. In the present study, we examined a group of 216(133 males) EV71-infected HFMD patients aged 1 to 6.7 years. There were 126 mild cases and 90 severe cases, in which 41 cases exhibited encephalitis that led to one death. All patients had a temperature ≥37℃ at the time of admission. 2. The genotype distributions and the allele frequencies of OAS1 rs1131454 were not different between healthy controls and EV71-infected patients. 3. When the mild and severe cases of HFMD were compared, the differences in genotype, allele, and carriage frequencies of OAS1 rs1131454 were significantly different. 4. Significant differences were observed in patients with EV71 infection in different genotype with respect to age and duration of fever. C-reactive protein(CRP) levels, white blood cell(WBC) counts, and blood glucose(BG) concentrations were significantly different for different genotypes. No significant differences were observed in gender, and the levels of alanine aminotransferase(ALT), aspartate aminotransferase(AST) among the genotypes. 5. CSF was measured for 41 EV71 encephalitis patients, and it was found that there were no significant differences in glucose, chloride, protein cell counts, and lymphocyte and monocyte counts of CSF according to genotype distributions of OAS1 rs1131454.Conclusion 1. In this study, we report an association between OAS1 polymorphism and susceptibility to severity of EV71-infection in a Chinese population. But in EV71-infected patients, a significantly higher frequency of A allele in OAS1 rs1131454 was observed in the severe cases. 2. The polymorphism of OAS1 rs1131454 A/G is not associated with the susceptibility to EV71 infection.
