Study On Relationship Between Single Nucleotide Polymorphism Of IL-10/TNF-α And The Susceptibility To Encephalitis Of Enterovirus71Infection

Objective:To investigate the severity of enterovirus EV71infection and genetic susceptibility factors of enterovirus71(EV71)infection by studying the SNP of InterleukinlO,Tumor necrosis factor a and the genetic susceptibility factors of EV71infection.Methods:103causes who were diagnosed Enterovirus EV71infection were selected as study group at Qingdao of Shandong.On the basis of the severity of the disease,The study group was divided into two groups,which were mild and critical case group respectively.Genomic DNA of white blood cell were extracted. then iMLDR was used to determine genotype for the C/A polymorphism at-592position of IL-10gene And the G/A polymorphism at-308position of TNF-a gene.Results:In EV71infection critical group, the A allele frequency of IL-10-592position was significantly higher than that in mild case group (30.4%vs.10.0%, OR3.923,95%CI1.798-8.561, P<0.05).Compared with that of the normal children. there were obvious statistical significance in EV71infection patients of A allele frequency of TNF-α-308position(10.7%vs.6.0%, OR2.033,95%CI1.027～4.025, P<0.05)and also there were obvious statistical significance between mild and critical case groups (17.9%vs.6.40%, OR3.406,95%CI1.304～8.896, P<0.05)Conclusions:In our studies,it suggests that the A allele of IL-10-592position and TNF-α-308position might contribute susceptibility to encephalitis of enterovirus71infection.TNF-a-308GG is the protective genotype which protects children keep away from EV71.
