Objective: The effect of Human bone marrow stem cells’ microfilaments structures transfected by h IGF-1(human insulin-like growth factor-1) inducing and aggrecanse-1(proteoglycans polymer enzyme-1) gene silencing.Methods: Used genetic recombination technique to build the lentiviral vectors with h IGF-1 overexpressing and aggrecanse-1 gene silencing. Transfected human bone marrow stem cells and marked as gene inducing group, gene silencing group and gene inducingsilencing group. The Human bone marrow stem cells transfected with lentivirus without gene served as control group 1, and the human bone marrow stem cells without treated served as control group 2. After transfected 7 days, detected gene expression with RTPCR. Using laser scanning confocal microscope observe microfilaments structures and measuring the average fluorescence intensity of microfilaments structure.Results: The fluorescence intensity of gene inducing-silencing group is higher than gene inducing and gene silencing group, gene inducing and gene silencing group are higher than control group 1and control group 2(P<0.01).And there were no difference between control group 1 and control group 2(P>0.05).Conclusion: Both gene inducing and gene silencing group promote microfilaments structures generation. The effect of gene inducing and silencing is more pronounced.
