Objective: To observe the antitumor effect and mechanism of recombinant human endostatin(Endostar) injection in tumor combined with radiotherapy on MFC gastric carcinoma xenografts in nude mice and the expression level of SDF-1/CXCR4、MMP-2and MMP-9.Methods: Xenograft mice modle of MFC gastric carcinoma were established.The mice were divided into 4 groups randomly,they are the endostar combined with radiotherapy group,the endostar group,the radiotherapy group,the control group.The largest diameter and the vertical diameter of tumor were measured at different time points to evaluated the antitumor effect.At the 15 th day, mice were executed.Reverse Transcription-Polymerase Chain Reaction( RT-PCR) was used to determine the expression of the SDF-1/CXCR4、MMP-2、MMP-9m RNA in each group.Results:The combination group significantly enhanced the effects of tumor growth inhibition,the rate of tumor inhibition in combined treatment group was 48.9%,while radiotherpy group was 33.0%,and Endostar group was 12.1%. Meanwhile SDF-1 、CXCR4 、 MMP-2 、 MMP-9 m RNA expression levels in the combination group was0.68±0.12,0.72±0.16,0.54±0.12,0.62±0.13,which was lower than in other groups and the difference was considered statistically significant(P<0.05).Conclusion: Endostar combined with radiotherapy can significantly ihibitor the growth of carcinoma,the mechnism may by downregulating the SDF-1/CXCR4、MMP-2、MMP-9, inhibiting tumor cell growth, invasion and metastasis.
