Objective:The aim of the study was to research the effects of radiotherapy and endostar on the tumor immune micro-environment of transplantable model of gastric carcinoma in mice.It explored the mechanism of action of tumor micro-environment of adjuvant radiotherapy and endostar.Methods:Establish thirty-two tumor-bearing mice models. They were divided into four group randomly,including control group,ES group,RT group and ES+RT group. Respectively starting from the 14 th day,every 2 days we tested tumor tissue maximum diameter(a) and minimum diameter(b) of tumor-bearing mice,then calculated tumor volume and the average, drown the tumor growth curve and calculated the inhibitory rate.In the 28 days,we killed all mice.Then we detected the proportion of MDSC、Treg Foxp3 of tumor tissue in tumor-bearing mice by flow cytometry and expression of cell factors such as IL-10 、 TGF-β1 by ELISA.We analysed the simple effect and the interaction of the two methods of treatment of endostar and radiotherapy with 2×2 factors factorial experiment design in this studies.Results: For control group,ES group,RT group and ES+RT group, the velocity of increase is less than that of the control groups, among which the combined treat group of Endostar and radiotherapy witnesses the slowest bulk velocity and the smallest bulk and has the greatest inhibitory rate; The results of variance analysis of 2*2 factorial designs indicate that the ES+RT group has the interaction effect in tumor inhibition and content reduction of IL-10、TGF-β1, and proportion decrease of MDSC、Treg Foxp3(P<0.05).It was positive effect.The Endostar and radiotherapy have the interaction effect. When radiotherapy is used regularly or not used at all, the use of Endostar witnesses the weight decrease of transplantation tumor and content reduction of IL-10、TGF-β1, and proportion decrease of MDSC 、 Treg Foxp3, compared with completely no use of Endostar(P <0.05).When Endostar is used regularly or not used at all, the use of radiotherapy witnesses the weight decrease of transplantation tumor and content reduction of IL-10、TGF-β1, and proportion decrease of MDSC 、 Treg Foxp3, compared with completely no use of radiotherapy. The difference was statistically significant(P<0.05).Conclusion:A possible mechanism of endostar combined with radiotherapy maybe through relieving immunosuppressive effect of tumor micro-environment,thus more obviously inhibit tumor growth.
