Curative effect, low toxic and new structural compounds were got from natural products, which could be used for drug research.This paper was divided into two parts: 1. The separation, purification, structure conformation of chemical constituents of Garcinia mangostana.The pericarps of Garcinia mangostana were collected in Shanghai of China in April 2014, and were identified by Associate Professor Zhangjun Cao, powdered and extracted with 95% ethanol. Then the resulting mixture extracted with acetic ether, which was isolated by repeated Column Chromatography(CC) on sillca gel, RP-18 and Sephadex LH-20, as well as by Preparative Thin Layer Chromatography(PTLC), High Performance Liquid Chromatography(HPLC) and recrystallization. From Garcinia mangostana, 22 compounds were isolated. The structures of these compounds were elucidated by means of spectroscopic methods including UV、ESI-MS、1D-NMR( H-NMR, C-NMR) and 2D(HMBC, HSQC), and 17 compounds were identified by spectral analysis: 1 13S-1:α-MangostinS-2:β-MangostinS-3: γ-MangostinS-4: 6-methoxy-tetrahydrothwaitesixanthoneS-5: Garcinone DS-6: GartaninS-7: Garcinone ES-8: MangostanateS-9: 6-methoxy-γ-mangostinS-10: 8-DeoxygartaninS-11: 11-Hydroxy-1-isomangostinS-12: AllantoinS-13: Stearic AcidS-14: Palmitic AcidS-15: StigmasterolS-16: RutinS-17: Single crystalAmong them, S-8、S-17 were the new compounds, S-12 was isolated from Garcinia Linn. for the first time, and S-13、S-14、S-15 were isolated from this plant for the first time, α-Mangostin is the main component of the pericarps of Garcinia mangostana. It is about 1%. 2. Pharmacological action research of Garcinia mangostana.All the isolated compounds were tested for their antioxidant activity(IC50) with DPPH(2,2-Diphenyl-1-(2,4,6-trinitrophenyl)hydrazyl) compared with vitamin C. Five compounds showed antioxidant activity : α-Mangostin, γ-Mangostin, Garcinone D, Gartanin, 6-methoxy-tetrahydrothwaitesixanthone. The antitumor activity in vitro of some compounds was evaluated by standard MTT(3-(4,5)-dimethylthiahiazo(-z-y1)-3,5-di-phenytetrazoliumromide)assay. We selected two kinds of tumor cell lines: human colon cancer cells(HCT116) and human liver cancer cells(Hep G2). The results showed that α-Mangostin, β-Mangostin, γ-Mangostin, Garcinone D and Gartanin exhibited the higher activity on cancer cells HCT116 and Hep G2.
