ObjectiveEsophageal squamous cell carcinoma(ESCC)is a common malignancy of the digestive system, Currently, there are limited clinical approaches for the early diagnosis and treatment of ESCC, resulting in a 10% five-year survival rate for patients. However, the full repertoire of genomic events leading to the pathogenesis of ESCC remains unclear. It is generally believed esophageal squamous cell carcinoma is the result of multi-factor, multi-gene, multi-stage synergy. It is widely concerned that some genes have important role in the occurrence and development.As a novel therapeutic targets, PSMD2 harbouring potentially drugable alterations. This study aimed to determine the role of PSMD2 on early onset of esophageal squamous cell carcinoma, our research will contribute to the early diagnosis and intervention of ESCC. Methods1. 55 cases of esophageal epithelium tissues which from department of Gastroenterology in Beijing Friendship Hospital, from 2010- 2015, with 10% formalin-fixed, paraffin-embedded. The specimens were taken from endoscopic submucosal dissection operation. 35 cases of men, 20 cases of women, aged from 43~81 years, the average age of 62 years. According to the Vienna Classification(2002), normal esophageal mucosa, low-grade intraepithelial neoplasia, High- grade neoplasia(intraepithelialor intramucosal) can be seen in the same tissue.2. Esophageal squamous cell carcinoma and matched adjacent tissues chip samples 69 cases.All cases were pathologically confirmed and collect clinical data.3. Using chi-square test analysis the differential expression of PSMD2 protein in normal esophageal mucosa, low-grade intraepithelial neoplasia, High- grade neoplasia and ESCC, adjacent tissues, P <0.05 was considered statistically determined difference. ResultsImmunohistochemical staining results showed that: PSMD2 protein is expressed normal esophageal mucosa, low-grade intraepithelial neoplasia and High- grade neoplasia, The positive rates are1.8%(1/55)、76.4%(42/55)、96.4%(53/55). the difference was statisically signifcant(P <0.05).PSMD2 protein is expressed ESCC, adjacent tissues, The positive rates are 71.01%(49/69),10.14(7/69). the difference was statisically signifcant(P <0.05). ConclusionsPSMD2 protein may be important a kind of bio-markers in the aspects of occurence in esophageal and provide new treatment ideas.
