| Objective To research the expression of DDX3 gene in advanced gastric cancer tissue and para-carcinoma tissue, analyzing the relationship between DDX3 gene and clinical pathological features of gastric cancer. To explore the possible function of DDX3 in gastric cancer development and metastasis.Method Sixty gastric cancer specimens and para-carcinoma tissue samples were obtained. Immunohistochemistry was used to detect the DDX3 protein level and RT-PCR was used to detect the DDX3 m RNA level. Then we compare the expression differences of DDX3 protein and DDX3 m RNA in carcinoma tissue and paracarcinoma tissue, and statistically analyze DDX3 expression with clinicopathologic features. Results The positive expression rate of DDX3 protein in gastric cancer tissue is 54.39%(31/60), and it is significantly higher than that of para-carcinoma tissue 6.67%(4/60), which is statistically significant(P=0.00).The DDX3 m RNA average absorbance value is higher in gastric cancer tissue(1.54±0.24) than those in para-carcinoma tissue(1.00±0.16)(P<0.05). In gastric cancer, the expression of DDX3 is uncorrelated with gender, age, tumor size and is related to the differentiation grades, invasion depth, lymph node metastasis, vascular invasion and TNM staging, and its expression level is rised along with the tumor progress(P<0.05).Conclusion The expressions of DDX3 gene are detected both in gastric cancer tissue and para-carcinoma tissue, but the expression in gastric cancer tissue is higher than para-carcinoma tissue, indicating that DDX3 might participate in the progress of gastric cancer. The high expression of DDX3 is related to the differentiation grades invasion depth, lymph node metastasis, vascular invasion and TNM staging and it may play a certain role in the invasion and metastasis of gastric cancer. |
