Objective:To explore the effect of Golgi protein 73(GP73) silencing via RNA interference(RNAi) on the invasion and migration ability of human hepatoma cells, and explain the mechanism to provide a theoretical basis for the hepatocellular carcinoma treatment of genic target GP73.Methods: A pair of designed and synthesized GP73 small interfering RNA(siRNA) was instantly transfected into HepG2 cells and SMMC7721 cells via cathodolyte liposome transfection method. Real time quantitative PCR(qRT-PCR) and ELISA were used to detect the GP73 expression at mRNA levels and protein levels in HepG2 cells and SMMC7721 cells, and the invasion and migration ability of hepatoma cells after transfection were separately observed by Transwell invasion assay, Transwell migration assay and wound healing assay. qRT-PCR and Western blot were used to measure the matrix metalloproteinase 2(MMP-2), MMP-9, E-cadherin, Snail and Vimentin expression at mRNA levels and protein levels in Hep G2 cells and SMMC7721 cells.Results: GP73 siRNA interference significantly down-regulated the GP73 expression at mRNA and protein levels in HepG2 cells and SMMC7721 cells, and knock-down of GP73 suppressed the invasion and migration ability. Expressions of Snail, Vimentin, MMP-9 and MMP-2 were significantly reduced by GP73 siRNA interference, but the expression of E-cadherin was increased.Conclusion: siRNA interference-mediated GP73 silencing can inhibit the invasion and migration ability of human hepatoma cells, and the possible mechanism may be related to the down-regulation of MMP-2 and MMP-9, and also may be related to the signaling pathway of Snail/E-cadherin/Vimentin.
