Objective:To examine the correlation between genetic polymorphisms(MS4A2 rs1441586 and FCER1 G rs11587213)of high-affinity IgE receptor( FcεRI) promoter region and penicillin allergy.Methods:A case-control method was used in the study, 210 healthy controls and 200 patients with penicillin allergy were selected,they were all the Han population in Gansu province, using the polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) to determine the distributions of MS4A2 rs1441586 and FCER1 G rs11587213 genetype and allele frequency. Sequencing was used to verify the results. Hardy-Weinberg test was choosed to determine whether the sample has representation, software of SPSS 21.0 was used in the statistical analysis, the different results in both groups were determined by the chi-square test.Results:There is no statistical differences between the allergy and control groups in the TT、TC and CC genotype frequencies of MS4A2 rs1441586(P=0.064),but the T、C allele frequencies in the two groups have significant statistical differences( P=0.048, OR=1.221, 95%CI =1.001-1.489); according to the gender groups, there is no statistical differences nother in the allele frequencies nor in the genotype frequencies;when stratified by age, the TC genotype frequency was different in non-aged patients between the two groups(P =0.009,OR=1.416,95%CI =1.088-1.842). There is no statistical differences between the allergy and control groups in the AA、AG and GG genotype frequencies of FCER1 G rs11587213, the frequency distribution of A、G in the two groups has statistically significant differences(P=0.028,OR=1.960,95%CI =1.063-3.614), the G allele plays particularly prominent role in males between the two groups when stratified by gender(P=0.038, OR=3.082,95%CI = 0.999-9.512); after the grouping by age, the A、G allele frequencies in elderly between the two groups have significant statistical differences(P =0.018,OR=2.970,95%CI =1.133-7.785),and the AG genotype frequencie also have significant statistical differences(P =0.002,OR=4.875,95%CI =1.502-15.827).Conclusion:The genetic polymorphisms of FcεRI promoter region possibility correlated to the penicillin allergy reaction, MS4A2 rs1441586 is associated with non-aged patients and FcεR1G rs11587213 is highly associated with the elderly and male patients.
