Objective:To explore the effects of Icotinib combined with CIK on the inhibits proliferation and induces apoptosis of various human lung adenocarcinoma cells. To provide the experimental basis for further research of Icotinib combined with CIK in the treatment of lung cancer. Methods:The inhibitory effect of Icotinib on HCC827 and A549 cells was detected by LDH assay and Cell Counting kit-8(CCK-8). The inhibitory effect of Icotinib combined with CIK on the growth of HCC827 and A549 cells was detected by CCK-8 and flow cytometry with Annexin V/PI staining. The effect of Icotinib on CIK cell subsets was detected by flow cytometry. Results:The inhibition rate of Icotinib to HCC827 and A549 increased with the increase of concentration(P<0.05). And more sensitive to HCC827(P<0.05). The killing effect of CIK cells on A549 and HCC827 were not different(P>0.05). The lethal effect increased with the increase of the effect/ target ratio.When combinedwith Icotinib, the effect of CIK was stronger, and the cell phenotypes or cytokine secretion was not affected(P>0.05). Conclusion:Icotinib was more sensitive to EGFR mutant lung adenocarcinoma cells. The EGFR mutation status had no effect on the killing effect of CIK cells. The combined effect of Icotinib and CIK on tumor cells was more effective. It is proved that Icotinib combined with CIK has a synergistic anti-tumor effect. And Icotinib had no effect on the phenotype of CIK cells.
