Objective :To investigate the VEGF gene transfected endothelial progenitor cell transplantation on high-fat hyperlipidemia rat penile corpus cavernosum local vascular regeneration,endothelial function in repair and erectile function of the influence.Methods :Choose mature Wistar rats, by feeding high fat forage and made of animal model of hyperlipidemia and endothelial function testing and drug erectile experiment confirmed to the animal model of ED 15, according to the weight divided into three groups: group I control group, II Group EPCs transplantation group, III group VEGF gene transfected EPCs transplantation group. Observed and compared by adenovirus will vascular endothelial growth factor gene transfected endothelial progenitor cell transplantation in hyperlipidemia rat penile corpus cavernosum, by HE staining and immunohistochemistry of detection of penile cavernous vascular density, RT-PCR and ELIASA method detection cavernous local VEGF expression and drug erectile experiment(artificial injection of apomorphine), and to explore the VEGF gene transfected endothelial progenitor cell transplantation on high-fat hyperlipidemia rat penile corpus cavernosum local vascular regeneration, endothelial function in repair and erectile function of the influence.Results :(1) HE staining and immunohistochemistry showed that: Group III and group II than group I vascular number of proliferated obviously. The difference(P < 0.01), with statistical significance;(2) ELIASA reverse transcription polymerase chain reaction(RT-PCR) and assay showed: Group III rats cavernosal local peripheral blood VEGF protein content is higher than that of group I and group II, the difference was statistically significant(P < 0.01) and group II rats cavernosal local peripheral blood VEGF protein was slightly higher than that of group I;(3) Artificial erection experiments can be obviously found that the rats in group III erectile times greater than in group II and group I, the difference has statistical significance(P < 0.05);(4) Intercellular adhesion molecule-1 in the blank control group was significantly higher than that in the other two groups, and the EPCs transplantation group was statistically significant(P < 0.05). There was no significant difference in the content of VEGF transfection group compared with that of the EPCs group(P > 0.05).Conclusion :VEGF gene transfected endothelial progenitor cell transplantation can promote vascular ed rat penile corpus cavernosum arteries endothelial function quick fixes, promote blood vessel regeneration, improve the penile hemodynamics and erectile function.
