Objective:Discuss(-)- Epigallocatechin gallate(EGCG) on gastric cancer(AGS) cell proliferation, apoptosis, as well as to the Sonic hedgehog(SHH) signaling pathway related protein expression of Ptch, Gli- 1.Methods:AGS cell lines at 37 ℃ and 5% CO2 saturated humidity temperature box for culture,changing fluid and batches.Experimental EGCG concentration of 25μg/ml, 50μg/ml,100 μ g/ml, and set the negative control group, the logarithmic phase cell during the test.Determined by MTT method and flow cytometry is used to test EGCG to AGSgastric cancer cell proliferation and apoptosis, cell climbing piece of immunohistochemical detection of SHH signaling pathways related protein expression of Ptch, Gli- 1.Results:EGCG intervention AGS cells after 48 h, as EGCG concentration increases, the decrease in the number of cells to stick wall became round, and form the floating populations, distribution of thin, visible to the cell after the disintegration of fragments,granules, etc;And the negative control group AGS cells stick wall is good, clear outline.As EGCG concentration increased, the AGE gastric cancer cell proliferation inhibition rate, apoptosis rate increased significantly(P<0.05);Increased with the concentration of EGCG, Gli-1 expression significantly reduced and a significant rise in Ptch expression(P < 0.05).Conclusion:EGCG could inhibit SHH signaling pathways related proteins to AGS gastric cancer cell apoptosis.
