Objective: to observe the expression and mechanism of HIF-1α and p53 protein at the altitude of 5000 m plateau hypoxia environment in SD rats, objective to investigate the protective effect of Astragalus Injection on the high altitude retinopathy.Methods: according to the principle of random group the 90 SD rats divided into the normal saline group, Astragalus injection group, Rhodiola injection group, the number of each group is 30, normal saline group rats by intraperitoneal injection with saline injection(15 ml/kg) before 30 minutes into the plateau environment simulation cabin, Astragalus group rats by intraperitoneal injection of Astragalus injection(15ml/kg) before 30 minutes into the plateau environment simulation cabin,Rhodiola group rats by intraperitoneal injection of Rhodiola injection(15 ml/kg) before 30 minutes into the plateau environment simulation cabin, after injection 0.5h each groph randomly selected 6 rats into the plateau environent cabin for 2h, 6h, 8h, 12 h, 24 h, to hold simulated altitude of 5000 m(11.3 kPa). When get out the rats after executed immediately remove eyes, then observed the changes HE staining in the rat retina changes, using the immunohistochemical method(IHC) different time periods in the rat retina tissue hypoxia inducing factor 1 alpha alpha(HIF- 1) and the changes of p53 gene expression, using the Image Pro Plus Image analysis software calculates the average absorbance positive cells [A] value, and analysis of the correlation of HIF-1alpha and p53.Results: 1. Each group rats pathological results: the high elevation hypoxia early,thickening of the inner retina edema of rats, shrinking over time increase the retinal nerve fiber layer, ganglion cells swelling degeneration, disordered arrangement of level. Astragalus injection group, rhodiola injection group retinal tissue morphological changes similar to normal saline group, reduced numbers of cells and retinal thickness thinning extent improve than physiological saline control group.2. Each group in the rat retina HIF-1 alpha, p53, test results: normal saline group were in the high altitude hypoxia 2h HIF-1 alpha, the expression of p53, gradually increase with time, expressed in 12 h enhanced obviously, 24 h express to peak(P < 0.05), theretina of HIF- 1 alpha, the expression of p53, more layer; Compared with normal saline group, Astragalus injection group and rhodiola injection group HIF-1alpha, p53 expression are reduced, statistically significant difference(P<0.05,P<0.05),Astragalus injection group of each time period HIF-1 alpha, the expression of p53 were slightly higher than the group of each time period expression, rhodiola injection group rosea HIF-1 alpha and expression of p53, correlation analysis results showed that both were positively correlated.(r- 0.460, P < 0.05)Conclusion:1. Astragalus injection can improve the damage degree of high altitude hypoxia rats retinopathy;2. Astragalus injection has a protective effect on retinal injury in rats with high altitude hypoxia, and its mechanism may be related to inhibiting the expression of HIF-1 and p53 factors in the retina.
