Objective: To study the effect of unilateral ureteral complete obstruction on the a poptosis of renal tubular epithelial cells and the expression of HO-1 and b FGF in r enal tubules.Methods: 80 SD rats were randomly divided into experimental group and control group. The former were randomly divided into 3, 5 and 7 days group of ureteral obstruction, 20 SD rats in each group. With 0.01% sodium pentobarbital, anesthesia by intraperitoneal injection of 1ml / kg, from the ventral midline incision into the abdominal cavity and find the bladder, in its vicinity revealed the separation of un ilateral ureter, in the lower 1 /3 ureter cut and 1# thread were ligated to the distal and proximal end, incision suture continued feeding. In the control group, unilateral ureteral exposure was performed. After the 3rd, 5th and 7th day of the experiment al group operation, bilateral kidney of corresponding time group were removed, afte r the control group renal surgery 7 days,at the same time remove bilateral kidney, with 8% formaldehyde fixed stand-by. All specimens were stained with HE to obs erve the pathological changes of the cell morphology, TUNEL staining method was used to calculate the apoptosis rate of renal tubular epithelial cells. The expression of HO-1 and b FGF in renal tubules was detected by immunohistochemistry.Results: 80 SD rats were all satisfied with the experimental design, and there wa s no accidental death after operation. The experimental group with the prolongation of obstruction, ipsilateral kidney volume gradually increased, surface color becomes dark, texture becomes tough, ureter, renal pelvis dilatation, cortical thinning; HE st aining showed glomerular atrophy and hyalinization, renal tubular epithelial cells sh owed vacuolar degeneration, renal interstitial cells infiltration, myofibroblasts, diffuse hyperplasia; renal tubular epithelial cell apoptosis; Immunohistochemistry showed that the HO-1 and b FGF protein of rats in the obstructed kidney all expressed, negat ive control groups were no positive expression. HO-1, b FGF mainly expressed in th e renal tubules,after obstruction positive expression was significantly increased. With the prolongation of obstruction time, the expression of HO-1 expression decreased, b FGF positive expression enhanced, the differences were significant(P< 0.05). Cor relation analysis showed that: HO-1 expression and renal tubular epithelial cell apop tosis rate was negatively correlated, b FGF expression and renal tubular epithelial cel l apoptosis rate was positively correlated, correlation coefficient r test exists signific ant difference(P < 0.05). There was no significant change in the side of the kidne y volume. The apoptosis of renal tubular epithelial cells still exist, but the rate of apoptosis was relatively slow with the time of obstruction.Conclusion: After unilateral ureteral obstruction, obstruction side and on the obstr uction side found renal tubular epithelial cell apoptosis. With the prolongation of ob struction, apoptosis rate increased significantly, the obstruction side change relatively slowly. The expression intensity of the renal tubular HO-1 and b FGF in ureteral obstruction were related of obstruction duration. The expression intensity of HO-1 with the prolongation of obstruction and decreased was negatively correlated with the rate of apoptosis of renal tubular epithelial cells, may have a inhibition of cell apoptosis protective role in unilateral ureteral obstruction in the process. The expression intensity of b FGF with the obstruction time prolonged, and was positively correlated with renal tubular epithelial cell apoptosis rate, which promoted nephropathy renal tubular fibrosis demonstrated in the process of obstructive.BFGF and HO-1 may be involved in the atrophy of renal parenchymal cells and the regulation of renal tubular fibrosis in obstructive nephropathy.
