Objective:To investigate the association of single nucleotide polymorphism(SNP rs325400 and rs34851361) of MEF2 A gene on chromosome 15q26 in patients with coronary artery disease(CAD).Methods(1) The information of the two SNPs was searched in the National Center of Biology Information(NCBI). The primers were designed with the Primer Premier 5.0 and the Blast software on-line.(2) The genomic DNA fragment of the 90 patients with coronary artery disease diagnosed by coronary angiography and 130 normal healthy controls were amplified by PCR.(3) The genotypes of the two SNPs were tested with PCR-RFLP and nucleotide sequencing.(4) The frequencies of the genotypes and alleles were evaluated with SPSS17.0.Results:(1) Three genotypes(AA, AG and GG) of rs325400 in MEF2 A were detected. Both frequencies of the rs325400 genotypes between CAD group(AA(23.3%)、AG(42.2%)、GG(34.5%)) and the control group(AA(10.8%)、AG(29.2%)、GG(60.0%)) and the rs325400 alleles between CAD group(A(44.4%)、G(55.6%)) and the control group(A(31.2%)、G(68.8%)) are significantly different(χ~2=7.750,P=0.021;χ~2=8.098,P=0.004).(2) There was only the AA genotype detected in both groups about the rs34851361, which was not meet the Hard-Wenberg balance principle, so the data invalid.Conclusion:The present findings suggest that the SNP genetic polymorphism in rs325400 may relate to the development of the CAD in the West South Chinese Han population and the prevalence of the CAD with allele A may be higher.
